    The lymphatic system

The major lymphaticvessels and collectionsof lymph nodes.Antigens are capturedfrom a site of infection, and the draining lymphnode to which theseantigens are

transported and wherethe immune response isinitiated.

    Morphology of a lymph node

    A. Diagram of a lymph nodeillustrating the T and B cell rich zones and the routes ofentry of lymphocytes andantigen (shown captured bya dendritic cell).

    B. Microanatomy of a lymphnode depicting the route oflymph drainage from thesubcapsular sinus, throughfibroreticular cell conduits,to the perivenular channelaround the HEV.

    C.Lymph node illustrating T

    cell and B cell zones

Segregation of B cells and T cells in a lymphnode.

    A. The path by which naive Tand B lymphocytes migrate toareas of a lymph node. Thelymphocytes enter through anartery and reach an HEV, fromwhere naive lymphocytes aredrawn to different areas of thenode by chemokines and bindselectively to either cell type. Also shown is the migration ofdendritic cells, which pick upantigens from the sites ofantigen entry, enter throughafferent lymphatic vessels, andmigrate to the T cell rich areasof the node.

    B. The B lymphocytes (ingreen), are located in thefollicles; the T cells (in red), inthe parafollicular cortex.

    Morphology of the spleen.

    A. Diagram of the spleenillustrating T cell and B cellzones, which make up the whitepulp.

    B. A section of human spleenshowing a trabecular artery withadjacent periarteriolar lymphoidsheath and a lymphoid folliclewith a germinal center.Surrounding these areas is thered pulp, rich in vascularsinusoids.

    C. T cell and B cell zones in thespleen, shown in a cross-section of the region around anarteriole. T cells (red) in theperiarteriolar lymphoid sheathand B cells (green) in the follicle

Cellular components of the cutaneous immunesystem.

The major components ofthe cutaneous immunesystem shown in thisschematic diagram includekeratinocytes, Langerhanscells, and intraepidermallymphocytes, all located inthe epidermis, and Tlymphocytes andmacrophages, located in thedermis.

    The mucosal immune system

A.Diagram of the cellularcomponents of the mucosalimmune system in theintestine.

    B. Mucosal lymphoid tissue

in the human intestine.Similar aggregates oflymphoid tissue are foundthroughout thegastrointestinal tract and therespiratory tract.

Pathways of T lymphocyte recirculation. Naive T cells preferentially

leave the blood and enterlymph nodes across theHEVs.

Dendritic cells bearingantigen enter the lymphnode through lymphaticvessels. If the T cellsrecognize antigen, they areactivated, and they return tothe circulation through theefferent lymphatics and thethoracic duct, which emptiesinto the superior vena cava,

then into the heart, andultimately into the arterialcirculation.

Effector and memory T cellspreferentially leave the bloodand enter peripheral tissuesthrough venules at sites ofinflammation.

    Ligh endothelial venules.

    A. Light micrograph of an HEV in alymph node illustrating the tallendothelial cells. B. Expression of L-selectin ligand on HEVs, stained with aspecific antibody by theimmunoperoxidase technique. (Thelocation of the antibody is revealed bya brown reaction product of peroxidase, which is coupled to theantibody; see Appendix III for details.)The HEVs are abundant in the T cellzone of the lymph node. C. A bindingassay in which lymphocytes areincubated with frozen sections of alymph node. The lymphocytes (staineddark blue) bind selectively to HEVs. D.Scanning electron micrograph of anHEV with lymphocytes attached to theluminal surface of the endothelial cells.
