NANOSTRUCTURED POLYMERIC MATERIALS AND BIOMATERIALS FOR
RECOGNITION: THE "PHAGE DISPLAY" TECHNOLOGY APPLIED IN BIOANALYSIS
ABSTRACT
The search of new recognition elements for biosensor development is of outcome importance as they
have great impact on the sensitivity and specificity of the device. Epitope-mimicking peptides are a
very attractive alternative to overcome some of the limitations of competitive immunoassays. They
bind to the same antibody paratope as the antigen and elicit a similar antibody response, therefore
they can be used in replacement of synthetically labelled antigens in applications where the
conjugation of the target to a carrier molecule is challenging, or it can cause toxicity to the user. Several
in vitro display technologies have been reported for screening antibody and peptide libraries. In
essence they mimic the natural in vivo process of antibody production however, but by far the most
widely spread technique is phage display. We have explored recently the development of different
phage display libraries, including domain antibody, single-chain fragment variable (scFv) as wells as
peptide libraries, to discover new recognition elements for biosensor development. Novel binders for
different target analytes including mycotoxins and immunosuppressant drugs were selected screening
domain antibody and scFv repertoires, whereas a 12-mer peptide library was used to identify epitope
mimicking peptides, or mimotopes, for their application as hapten-conjugates in competitive
immunoassays for mycotoxin analysis.
Several approaches for mycotoxin analysis using the phage-displayed binders the will be reported in
this presentation: 1) Development of a novel microarray-based immunoassay for fumonisin B1
analysis, with a detection limit of 11.1 ng mL-1 and a dynamic range from 17.3 to 79.6 ng mL-1, based
on the synthetic peptide originating from a phage-displayed library]. 2) Construction and application
of a recombinant fusion protein of the fumonisin B1 mimotope and a fluorescent protein to the
development of a homogeneous fluorescence quenching immunoassay based on gold nanoparticles
(AuNPs) for the detection of the toxin without the need for a labelled secondary antibody. The
approaches reported herein demonstrates the applicability of epitope mimics to sensitive
immunoassays.

RESUMEN
La búsqueda de nuevos elementos de reconocimiento para el desarrollo de biosensores es de gran
importancia ya que tienen un gran impacto en la sensibilidad y especificidad del dispositivo. Los
péptidos que imitan epítopos o mimopéptidos son una alternativa muy atractiva para superar algunas
de las limitaciones de los inmunoensayos competitivos, ya que son capaces de unirse al paratopo del
anticuerpo provocando una respuesta inmunoanalítica similar al epítopo o hapteno. De hecho, pueden
utilizarse en sustitución de antígenos etiquetados (también epítopos o haptenos) cuya síntesis puede
ser complicada, costosa o incluso peligrosa, al tener que utilizarse, en algunos casos, sustancias tóxicas.
Existen diferentes tecnologías de producción de bibliotecas de anticuerpos y/o péptidos que, en
esencia, imitan su proceso natural de obtención. Entre ellas, la presentación de anticuerpos y péptidos
en fagos es la técnica que mayor interés ha despertado en los últimos años. Recientemente hemos
explorado la utilización de diferentes bibliotecas de fagos con presentación de péptidos, además de
anticuerpos de cadena sencilla de tipo scFV ó VH, para descubrir nuevos biomateriales específicos para
el desarrollo de biosensores. Por ejemplo, se han seleccionado péptidos de 12-mer que mimetizan a
micotoxinas y se han utilizado en inmunoensayos de competición para el análisis de dichas sustancias.
Concretamente, se ha desarrollado: 1) un inmunoensayo basado en microarrays para el análisis de
fumonisina B1, basado en el péptido sintético que fue seleccionado mediante phage-display. 2)
producción de una proteína recombinante de fusión, constituida por un mimopéptido de fumonisina
B1 y una proteína fluorescente, para el desarrollo de un inmunoensayo homogéneo y directo basado
en nanopartículas de oro (AuNPs) para la detección dicha toxina. Estas dos estrategias de biosensado
que aquí presentamos demuestran la gran utilidad del phage-display como herramienta para el
desarrollo de biosensores.

(1) Development of smart recognition elements based on new chemical, biomimetic or biological materials.
The main topics I worked with include: genetically engineered proteins and cells, molecularly imprinted polymers
(MIPs), supramolecular and nanostructured receptors and biopolymers. In 2000, the design and preparation of
MIPs for the analysis of antibiotics and their application in optosensing devices. Between 2000 and 2014: (i)
synthesis of MIP filaments and nano/microspheres; (ii) preparation of MIPs with functional monomers water-
compatible; (iii) inclusion of signaling monomers on MIPs and their use on the development of label-free optical
sensors. During my postdoctoral period at Tufts (2009-2011), I tackled my research interests to investigate new
biological and biomimetic systems to generate selective engineered biorecognition elements. Since 2011, we are
also successfully exploring the optosensing capabilities of differently shaped gold nanoparticles, coated with
selective imprinted polymers or bioreceptors. And since 2014, we are also producing engineered phages for
being used in optosensing devices for small molecules monitoring.
(2) Development of novel signal amplification strategies.
My contribution to this objective includes: (i) molecular design and synthesis of fluorescent probes (antibiotics,
recombinant proteins); (ii) synthesis and exploration of the suitability for sensing of novel fluorescent
nanostructured materials (Gold NPs, QDs, polymeric micro/nanoparticles); (iii) use of these materials combined
with optical techniques (e.g. TIRFM, FLIM, Simoa).
(3) Implementation of sensing platforms for determination of analytes and/or flow-through analysis.
From 2009 to 2011 I worked with Prof. David R. Walt, the inventor of the Fiber Optic Array Sensors and the
Founding Scientist of Illumina, Inc. and Quanterix Co. My expertise expanded by working on projects that
involved multiplexed detection of biomarkers and pathogens using randomly ordered arrays based on coded
microspheres and fiber-optic platforms. Thus, I am currently interested in using my expertise on microarray and
multianalyte sensors in combination with my ability to create new nanostructured sensing materials, and readout
mechanisms, to develop sensor platforms with the aim of encompassing a growing number of applications

ELECTROCHEMICAL BIOSENSORS: NEW TOOLS FOR FOOD SAFETY AND CANCER
DIAGNOSIS
ABSTRACT
The early detection of diseases of increasing incidence and prevalence such as cancer and food security
are two of the main challenges facing society today. It is now widely accepted that early diagnosis of
cancer drastically reduces mortality from this disease, which in its early stages can be effectively
treated in more than 90% of patients with the eight most common cancers. In the field of food safety
and, in order to contribute to maximum consumer satisfaction and protection, the rapid and reliable
determination of food allergens and frauds is essential.
In these two areas there is an urgent need to develop alternative methods to the conventional ones,
capable of carrying out fast and multiple determinations with high sensitivity and selectivity and using
simple, low-cost and easily miniaturized instruments, which allow decentralized analysis in a routine
way in complex samples with minimum pre-treatments.
In response to this important demand, our research group has developed competitive electroanalytical
platforms for the sensitive, selective and rapid determination of biomarkers of different molecular
levels related to the presence of allergens and adulterations in food matrices and to the reliable and
early diagnosis of cancer in solid/liquid biopsies. Biosensing platforms are based on the use and
appropriate coupling of functionalized micromagnetic particles with new receptors, original bioassay
formats, novel amplification strategies and disposable electrochemical transducers for single/dual
detection.
In this talk the most relevant aspects of the bioplatforms developed for the determination of allergens
and adulterations of emerging relevance in foods (main allergens of egg, peanut, hazelnut, tomato and
adulterations with meat and milk of other animal species) and biomarkers related to epigenetic
(methylated nucleic acids and microRNAs) and/or metastases (extracellular and circulating protein
biomarkers) processes in cancers of high prevalence and mortality have been discussed.
The developed methodologies, which can be easily extended to the determination of other relevant
(bio)markers and integrated into multiplexed formats, have demonstrated their applicability in the
analysis of highly complex samples after minimum pre-treatments, providing results comparable to
those of other commonly used methodologies as opposed to those that prove to be advantageous in
terms of simplicity, time, cost of testing and the possibility of carrying out determinations in a
decentralized manner, characteristics that make them very attractive for their future implementation
in devices for use at the point of care for food safety and diagnosis, prognosis and clinical monitoring.

BIOSENSORES ELECTROQUÍMICOS: NUEVAS HERRAMIENTAS PARA SEGURIDAD
ALIMENTARIA Y DIAGNÓSTICO DE CÁNCER
RESUMEN
La detección temprana de enfermedades de creciente incidencia y prevalencia como el cáncer y la
seguridad alimentaria son dos de los principales retos que enfrenta la sociedad hoy en día. En la
actualidad está ampliamente aceptado que el diagnóstico temprano de cáncer reduce drásticamente
la mortalidad por esta enfermedad que en estadios tempranos puede tratarse eficientemente en más
del 90% de los pacientes que padecen los ocho tipos de cáncer más comunes.
En el campo de la seguridad alimentaria y, con el fin de contribuir a la máxima satisfacción y protección
al consumidor, es imprescindible la determinación rápida y fiable de alérgenos y fraudes alimentarios.
En estos dos ámbitos existe la necesidad urgente de desarrollar métodos alternativos a los
convencionales, capaces de realizar determinaciones rápidas y múltiples con elevadas sensibilidad y
selectividad y utilizando instrumentación de manejo sencillo, bajo coste y fácilmente miniaturizable,
que permitan realizar análisis descentralizados de forma rutinaria en muestras complejas con
pretratamientos mínimos.
En respuesta a esta importante demanda, se han desarrollado recientemente en nuestro grupo de
investigación distintas plataformas electroanalíticas competitivas para la determinación sensible,
selectiva y rápida de biomarcadores de diferente nivel molecular relacionados con la presencia de
alérgenos y adulteraciones en matrices alimentarias y con el diagnóstico fiable y temprano de cáncer
tanto en biopsias sólidas como líquidas. Estas plataformas biosensoras están basadas en el uso y
acoplamiento apropiado de partículas micromagnéticas funcionalizadas con nuevos receptores,
formatos originales de bioensayo, estrategias novedosas de amplificación y transductores desechables
electroquímicos para detección individual o dual.
Las bioplataformas desarrolladas, que pueden determinar (bio)marcadores de alérgenos y
adulteraciones de alimentos, relacionados con procesos epigenéticos y/o metastásicos en cánceres de
elevada prevalencia y mortalidad e integrarse en formatos de multiplexado, han demostrado su
aplicabilidad en el análisis de muestras de elevada complejidad tras pretratamientos mínimos,
proporcionando resultados comparables a los de otras metodologías de uso común frente a las que
demuestran ser ventajosas en términos de simplicidad, tiempo, coste de ensayo y posibilidad de
realizar determinaciones de forma descentralizada, características que hacen que resulten muy
atractivas para su futura implementación en dispositivos de uso en el punto de atención para seguridad
alimentaria y diagnóstico, pronóstico y seguimiento clínico.

Dra. en Química Analítica por la Universidad Complutense de Madrid desde 2004 donde en la actualidad es
Profesora Titular y pertenece al grupo de investigación de "Electroanálisis y (Bio)sensores electroquímicos"
dirigido por el Prof. José M. Pingarrón. 2008-2009: Contratada Juan de la Cierva en el CIB-CSIC. Enero 2010-Julio
2011: investigador visitante en el grupo del Prof. Joseph Wang de la UCSD (San Diego, USA). Sus investigaciones
se centran en el desarrollo de bioplataformas electroanalíticas enzimáticas, inmunosensoras y basadas en ácido
nucleicos para aplicaciones de vanguardia en los campos alimentario y clínico. Hasta la fecha ha (co)dirigido 8
Tesis doctorales y es (co)autora de 194 publicaciones (173 en JCR, 126 en Q1 y 50 en D1, índice h: 43), 19 capítulos
de libro, 8 patentes y 186 comunicaciones a Congresos. Desde Enero de 2019 es Editor Asistente de la revista
Electroanalysis (WILLEY-VCH).

DEVELOPMENT AND CHARACTERIZATION BY USING ANALYTICAL AND –OMIC
TECHNIQUES OF NOVEL HYBRID NANOSYSTEMS AGAINST CANCER AND
TUBERCULOSIS
ABSTRACT
Nanomedicine has recently emerged as a scientific discipline born from nanotechnology in
combination with the fields of engineering, physics, chemistry and biotechnology. In previous studies
our group demonstrated the ability of Se nanoparticles (SeNPs) to induce inhibition of the EiF3 protein
complex generally overexpressed in tumors, to arrest the cell cycle by partial inhibition of the Cdk1
protein and to inhibit tumor growth both in- vitro and in-vivo. Likewise, the ability of Ag nanoparticles
(AgNPs) to induce nucleolar stress in tumor cells was also demonstrated, which is one of the most
interesting therapeutic targets explored in recent years. Despite the results obtained and the
chemotherapeutic potential demonstrated by these two types of metallic NPs, there is a problem
related to their potential toxicity that could hinder its use at the systemic level. It is for this reason that
in we are currently working on their vehiculation through the development of new biocompatible
hybrid nanosystems, which allow the selective and highly efficient targeting of SeNPs and AgNPs to
tumor cells.
Regarding tuberculosis, the emergence of antibiotic-resistant Mtb strains constitutes a serious current
public health issue. For this reason, there is a growing interest in the development of new tuberculosis
vaccines, since the current licensed vaccine, Calmette-Guerin bacillus (BCG), provides limited
protection. A few years ago we demonstrated that Mtb actively secretes a series of proteins contained
in membrane-derived vesicles that are able to modulate the immune response. Therefore, these
natural vesicles could be used as vaccines, although their isolation is extremely complicated and
involves important problems of reproducibility. Based on this, we have designed nanosystems capable
of transporting these immunomodulatory proteins that would act as a preventive vaccine against TB.
In addition, since our group has also recently demonstrated the antibacterial capacity of AgNPs and
SeNPs against MTB, the development of a potential therapeutic vaccine based on a more complex
hybrid nanosystem, which in addition to having immunomodulatory proteins on its surface, it would
contain AgNPs or SeNPs that may act to reduce the bacterial load of infected cells, is also a part of our
current research line. The general strategy followed in this research is: (i) design and synthesis of the
nanosystems; (ii) evaluation of their stability, toxicity and localization in cells and tissues by
hyphenated atomic techniques; (iii) elucidation of the biomolecular mechanisms involved by
transcriptomics, proteomics and metabolomics.

ÓMICAS DE NUEVOS NANOSISTEMAS HÍBRIDOS COMO HERRAMIENTA FRENTE A
CÁNCER Y TUBERCULOSIS
RESUMEN
La nanomedicina, emergida como especialidad científica de la nanotecnología en combinación con los
campos de la ingeniería, física, química y biotecnología. En estudios previos nuestro grupo demostró
la capacidad de las nanopartículas de Se (SeNPs) para inducir la inhibición del complejo proteico EiF3
generalmente sobreexpresado en tumores, para arrestar el ciclo celular por inhibición parcial de la
proteína Cdk1 y para inhibir el crecimiento tumoral tanto in-vitro como in-vivo. De la misma manera,
también se demostró la capacidad de nanopartículas de Ag (AgNPs) para inducir stress nucleolar en
células tumorales, lo cual constituye una de las dianas terapéuticas más interesantes exploradas en los
últimos años. A pesar de los resultados obtenidos y al potencial quimioterapéutico demostrado por
estos dos tipos de NPs metálicas, existe un problema relacionado con su potencial toxicidad que podría
dificultar su uso a nivel sistémico. Es por ello, que actualmente estamos trabajando en su vehiculización
mediante el desarrollo de nuevos nanosistemas híbridos biocompatibles, que permitan dirigir las
SeNPs y AgNPs de forma selectiva y con alta eficacia a células tumorales.
En lo que se refiere a tuberculosis, la aparición de cepas de Mtb extremadamente resistentes a
antibióticos constituye un problema actual de salud pública muy importante. Por este motivo, existe
un interés creciente en el desarrollo de nuevas vacunas antituberculosas, ya que la vacuna licenciada
actual, bacillus Calmette-Guerin (BCG), proporciona una protección limitada. Hace unos años
demostramos en nuestro grupo que MTB secreta activamente una serie de proteínas contenidas en
vesículas derivadas de membrana que son capaces de modular la respuesta inmune. Por tanto, estas
vesículas naturales podrían ser usadas como vacunas, si bien su aislamiento es extremadamente
complicado y supone problemas importantes de reproducibilidad. En base a esto, hemos diseñado
nanosistemas capaces de transportar estas proteínas inmunomoduladoras que actuan como vacuna
preventiva frente a TB. Además, y dado que nuestro grupo también ha demostrado recientemente la
capacidad bactericida de AgNPs y SeNPs frente a Mtb, se está actualmente trabajando en el desarrollo
de una potencial vacuna terapéutica basada en un nanosistema híbrido más complejo, que además de
tener las proteínas inmunomoduladoras en su superficie, contenga AgNPs o SeNPs que actúan
reduciendo la carga bacteriana de células infectadas.
La estrategia utilizada: (i) diseño y síntesis de los nanosistemas; (ii) evaluación de su estabilidad,
toxicidad y localización en células y tejidos mediante técnicas atómicas acopladas; (iii) elucidación de
los mecanismos biomoleculares implicados mediante transcriptómica, proteómica y metabolómica.

STUDY OF THE NEPHROTOXICITY OF CISPLATIN AND NEPHROPROTECTIVE
STRATEGIES IN ANTITUMORAL THERAPIES BY MEANS OF ANALYTICAL-OMIC
TECHNIQUES.
ABSTRACT
Nephrotoxicity is the most limiting side-effect of cisplatin-based antitumor treatments. The use of
mass spectrometry (MS)-based -omics analytical tools have contributed to the understanding of the
complex biochemical processes involved in cisplatin-induced renal damage and the nephroprotective
effect of cilastatin. By using elemental imaging MS (LA-ICP-MS), cisplatin was proved to accumulate in
the proximal tubule renal cells (mainly in the S3 segment) by direct visualization of accumulated
platinum on rat kidney sections, leading to apoptotic cell death and loss of renal function. Co-
administration of cilastatin (an inhibitor of renal dehydropeptidase I) selectively protects from the
renal damage caused by cisplatin, blocking extrinsic apoptosis and impairing cell internalization
processes. This is also in agreement with the decrease in the cisplatin accumulation in renal tubule
cells during cilastatin co-administration, found by ICP-MS analysis. The combined use of elemental
(ICP-MS) and molecular MS with electrospray (ESI-MS) also helped to detect and elucidate the
platinated protein complexes generated upon treatment with cisplatin, which may contribute to its
cytotoxic effects.
Furthermore, MALDI-mass spectrometry imaging (MALDI-MSI) allowed detecting in rat kidney
sections, altered phospholipid levels in renal cortex and medulla connected to renal damage caused
by cisplatin. These levels tend to recover to those of control samples either totally or partially upon co-
administration of cilastatin. Multivariate methods allow classifying and discerning rat groups treated
with cisplatin presenting nephrotoxicity from control healthy groups and those administered with both
cisplatin and cilastatin, by using quantitative data from specific phospholipid variables. These altered
species observed during cisplatin-induced renal damage (PC, PE, PI, PS, LPLs, cardiolipins, etc.) might
be regarded as potential damage markers and could also facilitate the study of new nephroprotective
strategies.

ESTUDIO DE LA NEFROTOXICIDAD DE CISPLATINO Y DE ESTRATEGIAS
NEFROPROTECTORAS EN TERAPIAS ANTITUMORALES MEDIANTE TÉCNICAS
ANALÍTICAS –ÓMICAS
RESUMEN
La nefrotoxicidad es el efecto adverso más limitante de los tratamientos oncológicos basados en el
cisplatino. El empleo de herramientas analíticas -ómicas basadas en la espectrometría de masas (MS)
ha permitido comprender las complejas bases bioquímicas del daño renal generado por cisplatino y
estudiar el efecto nefroprotector de cilastatina. Mediante MS elemental de imagen (LA-ICP-MS) se
pudo comprobar que cisplatino se acumula en las células de túbulo proximal renal (principalmente en
el segmento S3) por visualización directa de Pt acumulado en secciones de riñón de rata, provocando
la muerte celular por apoptosis y la disfunción renal. La administración de cilastatina (inhibidor de la
deshidropeptidasa renal I) protege selectivamente del daño renal, bloqueando la vía extrínseca de la
apoptosis y evitando los procesos de internalización celular. Esto también se observó en una reducción
de la acumulación de cisplatino en las células de túbulo proximal detectada mediante ICP-MS.
Empleando la MS molecular con electrospray (ESI-MS) e ICP-MS también se logró detectar y elucidar
la formación de complejos proteicos platinados que aparecen durante la administración de cisplatino
y que pueden contribuir a sus efectos citotóxicos.
Además, la técnica MALDI-espectrometría de masas de imagen (MALDI-MSI) permitió detectar, en
secciones renales de rata, alteraciones de fosfolípidos de corteza y médula renales que aparecen
asociadas al daño renal causado por cisplatino y que tienden a mantenerse en los niveles de los
controles de forma total o parcial durante la co-administración de cilastatina. Mediante métodos
multivariantes se pudo clasificar y distinguir grupos de ratas tratadas con cisplatino presentando daño
renal, de grupos de ratas sanas control y protegidas con cilastatina, en base a la abundancia de
diferentes especies de fosfolípidos. Estas especies alteradas durante el daño renal (PC, PE, PI, PS, LPLs,
cardiolipinas, entre otras) podrían servir de marcadores de daño renal y facilitar el estudio de posibles
nuevos nefroprotectores.

AGRI-FOOD RESIDUES AS A VALUABLE SOURCE OF BIOACTIVE POLYPHENOLS
ABSTRACT
Agriculture by-products present, in general, low added values. However, they contain significant
amounts of bioactive compounds such as polyphenols. Therefore, these by-products could be
reintroduced in the pharmaceutical, cosmetic and food industries as preservatives in foods, as
ingredients in functional foods or even as natural dyes.
Polyphenols are natural compounds present in a wide variety of foods and beverages with beneficial
and healthy properties, mainly due to their protective effect against oxidative stress. In this research,
selective extraction of polyphenols was performed from the residues generated in the brewing, wine
making and fruit juice manufacturing industries, and in the waste generated during the coffee
beverage preparation, using extraction with low cost and toxicity solvents and/or solid phase matrix
dispersion (MSPD) with TiO2 nanoparticles. In addition, analytical methods were developed for
identification and quantification of individual extracted polyphenols, based on capillary LC-DAD and
HPLC-ESI-MS-MS. The results obtained were evaluated by chemometric tools such as multifactorial
ANOVA, experimental design and principal component analysis.
In the case of beer, it was observed that residual brewing yeast contained appreciable amounts of
polyphenols, because they were bio-absorbed during the fermentation process. Regarding citrus peels,
it was found that the peel nature affected the type and quantity of polyphenols extracted. All citrus
peels evaluated and, especially, clementine peels, were a considerable source of hesperidin, with
potential interest in the cosmetic industry for its photoprotective effect, presenting the lemon peel
the highest content of naringin. On the other hand, spent coffee grounds from different coffee
varieties were analyzed, observing for some varieties and specific polyphenols, higher contents in the
coffee waste extracts than in the coffee beverage.

OBTENCIÓN DE COMPUESTOS BIOACTIVOS A PARTIR DE RESIDUOS DE LA INDUSTRIA
AGROALIMENTARIA
RESUMEN
Los subproductos de la agricultura tienen, en general, un bajo valor añadido. Sin embargo, pueden
contener cantidades significativas de compuestos bioactivos como polifenoles. Por ello, la no
reutilización de estos residuos supone la pérdida de productos y un deterioro del medio ambiente.
Estos subproductos pueden ser reintroducidos en industria farmacéutica, cosmética y en la alimentaria
como conservantes en alimentos, ingredientes de alimentos funcionales o colorantes naturales.
Los polifenoles son compuestos naturales y poseen propiedades saludables y beneficiosas,
fundamentalmente por su efecto de protección frente al estrés oxidativo. Se encuentran presentes en
una gran variedad de alimentos y bebidas y han despertado gran interés desde el punto de vista
nutricional, por sus acciones no solo en el estado de salud, sino en la prevención de las alteraciones
funcionales y estructurales de diversas enfermedades.
Se ha llevado a cabo la extracción selectiva de polifenoles en residuos de las industrias cervecera,
vitivinícola y de fabricación de zumos de frutas, y en los residuos generados durante el proceso de
elaboración de la bebida de café, utilizando disolventes de bajo coste y baja toxicidad y/o dispersión
de matriz en fase sólida (MSPD) con nanopartículas de dióxido de titanio. También, se han desarrollado
métodos para la caracterización y cuantificación individual de los polifenoles extraídos mediante
técnicas cromatográficas; LC capilar-DAD y HPLC-ESI-MS-MS. Los resultados se han evaluado utilizando
herramientas quimiométricas como ANOVA multifactorial, diseño experimental y análisis de
componentes principales. Los extractos polifenólicos obtenidos se han caracterizado en términos de
parámetros globales como el contenido total de polifenoles y flavonoides y la capacidad antioxidante.
En el caso de la cerveza, se ha podido observar que tanto la levadura gastada como el sobrenadante
contienen cantidades apreciables de polifenoles si bien, hay más en la levadura porque los bioabsorbe
durante el proceso de fermentación. En el caso de las pieles de cítricos evaluadas, se ha constatado
que el tipo de piel condiciona la metodología analítica utilizada para la extracción y que afecta al tipo
y cantidad de los polifenoles extraídos. Todas las pieles de cítricos evaluadas, especialmente,
mandarina, son fuente de hesperidina, con interés en la industria cosmética por su efecto
fotoprotector, presentando la piel de limón el mayor contenido de naringina. En relación con los
residuos de café, se han analizado los posos de distintas variedades observándose, para algunas
variedades y polifenoles, mayores contenidos en los extractos procedentes de los residuos que en la
bebida del café (extracto acuoso).

ABSTRACT
No wonder nanomaterials present new or improved properties in comparison with bulk materials
because of their small size. These nanomaterials have a high surface to volume ratio, which means a
large proportion of accessible atoms in their structure. This fact is really interesting since it increases
the catalytic activity of the nanoscale materials allowing their use in wide applications such as
adsorption, (electro)catalysis, immobilization of biomolecules, biomedicine, food packaging,
capacitators and batteries, …
Besides that, a large number of synthetic routes allow the size and shape control of the nanomaterials
providing the design of these materials for the desired application.
In our group, novel nanomaterials are developed and combined to obtain hybrid nanomaterials that
present even better properties than the separated nanomaterials. These new materials are employed
in the development of electrochemical sensors, acting as transducers with improved features and
simplify the immobilization of the selected receptor.
Furthermore, we build smart nanomaterials able to respond by changing their mechanical, physical or
chemical properties, to external stimuli or their environment such as temperature, light or chemicals.
These nanomaterials based on mesoporous silica, controlled by enzymes and capped with responsive
molecular gates, are employed as control drug delivery systems.

RESUMEN
No es de extrañar que los nanomateriales presenten nuevas o mejoradas propiedades frente a estos
materiales a tamaño macroscópico. Estos nanomateriales presentan una gran relación superficie-
volumen, lo que implica una gran proporción de átomos accesibles en su estructura. Este hecho resulta
realmente interesante debido a que incrementa la actividad catalítica de los materiales en la
nanoescala, permitiendo su empleo en un amplio rango de aplicaciones como adsorción,
(electro)catálisis, inmovilización de biomoléculas, biomedicina, envasado de alimentos,
condensadores y baterías…
Además de esto, un gran número de rutas sintéticas permiten controlar el tamaño y morfología de
estos nanomateriales, posibilitando el diseño de estos materiales para la aplicación deseada.
En nuestro grupo, desarrollamos y combinamos novedosos nanomateriales para obtener
nanomateriales híbridos, que presentan incluso mejores propiedades que los nanomateriales por
separado. Estos nuevos nanomateriales se emplean en el Desarrollo de sensores electroquímicos,
actuando como transductores con rasgos mejorados y simplificando la inmovilización de los receptores
seleccionados.
Además, nosotros construimos nanomaterials inteligentes capaces de responder mediante un cambio
en sus propiedades mecánicas, físicas o químicas frente a estímulos externos o su ambiente, tales
como temperatura, luz o compuestos químicos. Estos nanomateriales basados en sílice mesoporosa,
controlados por enzimas y bloqueados con puertas moleculares sensibles, son empleados como
sistemas de entrega controlada de fármacos.

ZEBRAFISH LARVAE AS A MODEL OF ECOTOXICOLOGICAL BIOACCUMULATION
ABSTRACT
In recent years, promoted by some national and international agencies (OECD, US-EPA, REACH-EU, ...)
and seeking to reduce animal experimentation, research is being conducted on the use of alternative
testing methods, where the use of staging primary development (larvae, embryos, ...) of animals is
presented as one of the most viable alternatives.
Following this path, the research group to which I belong has been testing the use of zebrafish larvae
as an alternative to the use of adult fish for the calculation of bioaccumulation factors. Using the same
restrictive conditions of the OECD 305 test (temperature 26 ± 2ºC, dissolved oxygen ≥ 60% and pH 6-
8.5, concentration of the substance to be tested below 1% of its LC50 and stable during the whole
period of the experiment, with a maximum variation of 20%, a maximum concentration of organic
solvent of 0.1 mL-1) the accumulation period is replaced by 48 hours instead of the 28 days indicated
in the OECD 305 and 24 hours of purification instead of the 14 days indicated in the original test. For
each compound to be tested, 1000 zebrafish embryos of 72 hpf are manually selected.
This model, following the recommendations of a panel of experts in alternative tests that established
some guiding principles that should be followed to select the list of chemicals to test alternative
protocols for acute lethality tests, has been used for the calculation of bioconcentration factors of
organic, inorganic compounds, or new emerging contaminants such as nanoparticles or drugs.

LAS LARVAS DE PEZ CEBRA COMO MODELO DE ECOTOXICOLÓGICO DE

RESUMEN
En los últimos años impulsados por algunas agencias nacionales e internacionales (OCDE, US-EPA,
REACH-UE, …) y buscando reducir la experimentación con animales, se investiga en la utilización de
métodos de testado alternativos, en donde la utilización de estadios de desarrollo primarios (larvas,
embriones…) de animales se presenta como una de las alternativas más viables.
Siguiendo esta misma línea de investigación, el grupo de investigación al que pertenezco viene
testando la utilización de larvas de pez cebra como alternativa a la utilización de peces adultos para el
cálculo de factores de bioacumulación. Utilizando las mismas condiciones restrictivas del test OCDE
305 (Temperatura 26±2ºC, oxígeno disuelto ≥ 60% y pH 6-8.5, concentración de la sustancia a testar
por debajo del 1% de su LC50 y estable durante todo el periodo del experimento, con una variación
máxima del 20%, una concentración máxima de disolvente orgánico de 0.1 mL-1) se sustituye el
periodo de acumulación por 48 horas en vez los 28 días indicados en el OECD 305 y 24 horas de
depuración en vez de los 14 días indicados en el test original. Para cada compuesto a testar, se
seleccionan manualmente 1000 embriones de pez cebra de 72 hpf.
Dicho modelo, siguiendo las recomendaciones de un panel de expertos en pruebas alternativas que
estableció algunos principios rectores que deberían seguirse para seleccionar la lista de productos
químicos para probar protocolos alternativos para pruebas de letalidad aguda, se ha utilizado para el
cálculo de factores de bioconcentración tanto de compuesto orgánicos, inorgánicos, o nuevos
contaminantes emergentes como las nanopartículas o fármacos.

MATERIALS OF THE FUTURE, ARTIFICIAL ANTIBODIES: SYNTHESIS, DESIGN AND

ABSTRACT
Molecular imprinting technology has evolved a great deal along its 40 years of extensive application to
the synthesis of organic and hybrid materials. Conceptually, it was born to prepare synthetic materials
able of mimicking the selective binding of target molecules, which is characteristic of antibodies and
enzymes.
Molecularly imprinting allows the design and preparation of tailor-made polymer materials for
(selective) recognition of chemical species. Thereby, Molecularly Imprinted Polymers (MIPs) contain
specific recognition sites, with a shape and geometry of functional groups complementary to those
present in the template molecule. For a MIP preparation, the selected print molecule, namely the
analyte or a surrogate molecule, interacts through covalent or non-covalent bonds with functional
monomers that are polymerized in the presence of a cross-linker to form a three dimensional
structure. Upon template removal, the polymer will bear specific recognition sites with

materials have been successfully applied in several analytical techniques, such as solid-phase
extraction, liquid chromatography, capillary electrophoresis, capillary electrochromatography,
catalysis, and as selective sorbents in chemical sensors for the determination of several families of
chemical compounds, such as antibiotics, toxins, pesticides, heavy metals, peptides and proteins
among others.
In this lecture the author will present an overview of the evolution of the Molecular imprinting
technique, along the last decades and the perspectives concerning about the new synthesis formats
and applications for the future.

RESUMEN
La tecnología de impronta molecular ha evolucionado mucho a lo largo de los últimos 40 años de se
ha aplicado de forma intensiva como síntesis de materiales orgánicos e híbridos. Conceptualmente,
nació para preparar materiales sintéticos capaces de imitar la unión selectiva de moléculas diana, al
igual que anticuerpos y enzimas [1].
La impronta molecular permite el diseño y la preparación de materiales poliméricos a medida, para el
reconocimiento (selectivo) de especies químicas. Por lo tanto, los Polímeros Molecularmente
Impronytados (MIP) contienen sitios de reconocimiento específicos, con una forma y geometría de
grupos funcionales complementarios a los presentes en la molécula de la plantilla. Para una
preparación de un MIP, la molécula seleccionada, el analito o una molécula sustituta, interacciona
través de enlaces covalentes o no covalentes con monómeros funcionales que polimerizan en
presencia de un entrecruzante para formar una estructura tridimensional. Al retirar la plantilla, el
polímero contendrá sitios de reconocimiento específicos con tamaño, geometría y disposición
complementarios de los grupos funcionales al analito objetivo. Estos materiales se han aplicado con
éxito en varias técnicas analíticas, como extracción en fase sólida, cromatografía líquida, electroforesis
capilar, electrocromatografía capilar, catálisis y como absorbentes selectivos en sensores químicos
para la determinación de varias familias de compuestos químicos, como antibióticos, toxinas,
pesticidas, metales pesados, péptidos y proteínas entre otros [2,3]. En esta conferencia, el autor
presentará una descripción general de la evolución de la técnica de impresión molecular a lo largo de
las últimas décadas y las perspectivas sobre los nuevos formatos de síntesis y aplicaciones para el
futuro.

El autor de esta presentación ha dirigido 1 tesis doctoral, 4 proyectos fin de máster, 1 proyecto ERASMUS y ha

artículos científicos en revistas indexadas en el JCR, dos capítulos de libros, y es co-inventor de tres patentes. El
Dr. Urraca ha sido miembro de la junta directiva de la Sociedad de Espectroscopia Aplicada (2004-2008). Fue
finalista al premio mejor estudiante de tercer ciclo según los premios Lilly 2006. Fue premio extraordinario de
Doctorado por la facultad de Química de la UCM en el año 2007, recibió el premio a la mejor tesis doctoral por
la Sociedad de Espectroscopía Aplicada en el mismo año y Premio Arquímedes en el año 2013.

THE MULTIPLE FACES OF A GOLD NANOPARTICLE
ABSTRACT
Nanoplasmonics can be defined as the science that studies the excitation of surface plasmon
resonances on metal nanostructures, and their potential technological applications. Such localized
surface plasmons arise from the interaction between light and the conduction electrons in
nanostructured metals, yielding bright colours in disperse metal nanoparticles. These colours can be
tuned by changing different parameters such as the size, shape, and composition of the particles, but
also the dielectric nature of the surroundings. In the case of anisotropic particles, the optical response
directly depends on the polarization of the incident light, which allows an additional degree of
manipulation. Moreover, these localized surface plasmons give rise to high electric fields at the
metallic surface, which can be controlled and amplified from colloidal self-assembly. All these effects
open new perspectives for designing new devices with applications in a wide variety of fields.

RESUMEN
La nanoplasmónica se define como la ciencia que estudia la excitación de resonancias plasmónicas
superficiales localizadas en nanoestructuras metálicas y sus posibles aplicaciones tecnológicas. Dichos
plasmones superficiales localizados surgen de la interacción de la luz con los electrones de conducción
confinados en una nanoestructura de metal, originando colores brillantes en nanopartículas metálicas
dispersas. El color obtenido puede ser modificado a través de cambios en el tamaño, la forma o la
composición de las partículas, así como de la naturaleza dieléctrica de su entorno. En el caso de
partículas anisótropas, la respuesta óptica depende directamente de la polarización de la luz incidente,
lo cual permite un grado adicional de manipulación. Dichos plasmones superficiales originan campos
eléctricos de gran intensidad en la superficie metálica, que pueden ser controlados y amplificados a
través de ensamblajes coloidales. Todos estos efectos abren un gran abanico de posibilidades para
diseñar dispositivos con aplicaciones en campos muy diversos.

INTERACTION OF SURFACTANTS WITH POLYMERS AND NANOPARTICLES: FROM
KNOWLEDGE TO APPLICATION
ABSTRACT
The understanding of the physico-chemical bases underlying the interaction of polymers and
nanoparticles with surfactants presents important implications in different scientific and technological
aspects affecting the quality of life of modern society. An example of the interaction between polymers
and surfactants is found in the formulation of hair care products, where complex mixtures are used.
Shampoos and conditioners are widely used to clean and improve the sensorial properties of hair
fibers, with the application of such products relying in the deposition of polymers and polymer+
surfactant complexes on a rather large surface (c.a. 6m2∙g−1). In aqueous solution, polymers interact
with surfactants leading to the formation of soluble complexes. The choice of the proper combination
of physico-chemical variables allows one to obtain a single phase shampoo which undergo phase
separation upon dilution during shampooing process, leading to the deposition of conditioning agents
onto hair fibers. The concept of forming and depositing supramolecular complexes onto hair is
considered the main mechanism involved in the conditioning effect of cosmetic formulations.
However, little is known about the actual interactions and their influence upon the structure at the
interface that impart the observed lubricant effect. Other aspect in which the interaction of colloidal
systems with surfactants is involved is the toxic effect of nanoparticles upon inhalation. Environmental
pollutants are inhaled and transported along the respiratory tract until the alveoli where interact with
the lung surfactant layer, the modification of the physico-chemical properties of such layer due to the
incorporation of nanoparticles is the main driving force of the potential toxicity of nanoparticles. The
understanding of the effect of particles in the physico-chemical properties of lung surfactant layers
may provide useful insight for performing a preliminary evaluation of the toxicity of inhaled powders.

RESUMEN
La comprensión de las bases físico-químicas que subyacen a la interacción de polímeros y
nanopartículas con tensioactivos presenta importantes implicaciones en diferentes aspectos
científicos y tecnológicos que afectan la calidad de vida de la sociedad moderna. Un ejemplo de la
interacción entre polímeros y tensioactivos se encuentra formulación de productos para el cuidado
capilar, en la cual se utilizan mezclas de una alta complejidad. Los champús y acondicionadores se
utilizan para la limpieza y mejora de las propiedades sensoriales de las fibras capilares, estando la
aplicación de dichos productos basada en la deposición de polímeros y complejos polímero-
tensioactivo en una superficie bastante grande (aproximadamente 6 m2 g-1). En disolución acuosa, los
polímeros interactúan con los tensioactivos, resultando en la formación de complejos solubles. La
adecuada combinación de variables físico-químicas permite obtener un champú monofásico que sufre
una separación de fases al diluirse durante el proceso de lavado, llegándose como resultado a la
deposición de agentes acondicionadores sobre las fibras capilares. El concepto de formar y depositar
agregados supramoleculares sobre el cabello se considera el mecanismo principal involucrado en el
efecto acondicionador de las formulaciones cosméticas. Sin embargo, poco se conoce en relación a las
interacciones reales y su influencia sobre la estructura en la interfase que conduce al efecto lubricante.
Otro aspecto en el que se encuentra involucrada la interacción de sistemas coloidales con tensioactivos
es la toxicidad de nanopartículas inhaladas. Las contaminates ambientales son inhalados y
transportados a lo largo del tracto respiratorio hasta que en los alvéolos interactúan con la capa de
tensioactivo pulmonar, la modificación de las propiedades físico-químicas de dicha capa debido a la
incorporación de nanopartículas es la principal fuerza motriz de la potencial toxicidad de las
nanopartículas. La comprensión del efecto de las partículas en las propiedades físico-químicas de las
capas de tensioactivo pulmonar puede proporcionar información de utilidad para realizar una
evaluación preliminar de la toxicidad de los polvos inhalados.

BIOPHYSICS OF MITOCHONDRIAL MEMBRANES
ABSTRACT
Iván López Montero is the principal investigator of the Mitochondrial Membranes Lab (MML), a
member research team of the Biophys-hub (UCM) and the Research Institut Hospital Doce de Octubre.
The MML focuses on understanding the membrane processes occurring in mitochondria to identify
new therapeutic targets against mitochondrial diseases. Along with this main scientific objective, the
MML is currently developing three research lines using biophysical and biochemical approaches:
• Membrane biophysics of ATP synthase: Fabrication of artificial bio-inspired systems able to in
situ produce and release ATP. Important achievements have been recently published in Angewandte
and PNAS (1,2).
• Identification of new therapeutic targets based on mitochondrial membrane mechanics
against mitochondrial diseases associated with the mitochondrial fusion machinery and cancer. This
includes the study of the molecular mechanisms of mitochondrial fusion and the mitochondrial
membrane remodelling promoted by lipophilic cationic probes (3,4).
• Development of experimental tools for real time visualization of intracellular ATP (5).

RESUMEN
Iván López Monmtero es investigador principal del Laboratorio de Membranas Mitocondriales (LMM),
equipo vinculado al grupo de investigación Biophys-hub de la Universidad Complutense y también al
área temática de Enfermedades Raras y de Base Genética del Instituto de Investigación Hospital Doce
de Octubre. Desde su fundación en 2014, el LMM tiene como objetivo general el estudio de los
procesos de membrana que ocurren en la mitocondria para identificar nuevas dianas terapéuticas
contra enfermedades mitocondriales. Con este objetivo principal vertebrador, el LMM desarrolla
actualmente 3 líneas de investigación mediante el uso herramientas tanto bioquímicas como
biofísicas:
• Biofísica de membranas de la ATP sintasa. Fabricación de sistemas artificiales bioinspirados,
capaces de fabricar ATP in situ y liberarlo en el interior. El LMM ha conseguido importantes logros en
esta línea publicados recientemente (1,2).
• Identificación de nuevas dianas terapéuticas basadas en la mecánica de membranas
mitocondriales asociadas a la maquinaria de fusión mitocondrial y cáncer. Esto incluye el estudio de
los mecanismos moleculares de fusión mitocondrial y el remodelado de las membranas mitocondriales
promovido por sondas catiónicas lipofílicas (3,4).
• Diseño y síntesis de herramientas experimentales para la visualización en tiempo real de
metabolitos mitocondriales (5).

MOLECULAR SIMULATION OF GAS CLATHRATES
ABSTRACT
Clathrates are inclusion compounds in which a gas molecule is enclosed within a solid-state lattice.
Numerous types of clathrates depending upon the nature of the solid network are known. When the
solid lattice is formed by water molecules are named clathrate hydrates or gas hydrates. These
compounds are formed under high pressure and at moderately low temperatures. Their structure
consists of a three-dimensional framework of hydrogen bonded water molecules within which are
incorporated a small number of molecules, usually gas molecules, such as as CH4, C2H6, C3H8, iC4H10,
CO2, H2S, N2, O2, H2 and noble gases. Also, heavier hydrocarbons like tetrahydrofuran or cyclopentane
can form clathrate hydrates. Since their discovery, a large number of clathrate hydrate systems have
been studied for their scientific interest and their potential applications in many different fields such
as the energy sector or the environment. In view of the large number of applications, it is desirable to
understand the behavior of these compounds. In the last years, studies on clathrate hydrates have
been growing exponentially given their relevance. However, there are still many questions unresolved
in the study of these compounds, for instance, the mechanisms of formation/dissociation of clathrate
hydrates. Understanding these mechanisms at a fundamental level is crucial in the search for novel
applications and improvement existing ones. Currently, computer simulation has become in an
indispensable tool in many scientific disciplines providing a bridge between the microscopic properties
of a system and its macroscopic properties. Computer simulation in studies about capture and
separation of gases plays a key role giving a vision of problem at molecular level.

RESUMEN
Los clatratos de gas son compuestos de inclusión en los que una molécula de gas se aloja en el interior
de una red cristalina sólida. Dependiendo de la naturaleza de la red sólida se conocen numerosos tipos
de clatratos. Cuando la red sólida está formada por moléculas de agua se denominan hidratos de
clatrato o hidratos de gas. Estos compuestos se forman a alta presión y a temperaturas
moderadamente bajas. Su estructura consiste en un entramado tridimensional de moléculas de agua
unidas por enlaces de hidrógeno dentro de las cuales se incorporan un pequeño número de moléculas,
generalmente moléculas de gas, como CH4, C2H6, C3H8, iC4H10, CO2, H2S, N2, O2, H2 y gases nobles.
Además, hidrocarburos más pesados, como el tetrahidrofurano o el ciclopentano, también pueden
formar hidratos de gas. Desde su descubrimiento, se han estudiado una gran cantidad de sistemas de
hidratos de gas debido a su interés científico y a sus aplicaciones en diferentes campos, como el sector
energético o el medio ambiente. En los últimos años, dada su relevancia, los estudios sobre los hidratos
de gas han crecido de manera exponencial. Sin embargo, todavía hay muchas preguntas sin resolver
en el estudio de estos compuestos, como, por ejemplo, los mecanismos de formación / disociación de
los hidratos de gas. Comprender estos mecanismos a un nivel fundamental resulta crucial en la
búsqueda de nuevas aplicaciones y en la mejora de las ya existentes. Actualmente, la simulación por
ordenador se ha convertido en una herramienta indispensable en muchas disciplinas científicas ya que
proporciona un puente entre las propiedades microscópicas de un sistema y sus propiedades
macroscópicas. La simulación por ordenador en los estudios sobre captura y separación de gases juega
un papel clave en estos sistemas ofreciendo una visión única del problema a nivel molecular.

GIANT VESICLES AS SWIMMERS AT LOW REYNOLDS NUMBER
ABSTRACT
Some biological microorganisms crawl or swim in the absence of external appendages thanks to the
coordinated motion of organelles within the outer membrane, which push the cells forward through
intracellular forces. In our work, we remotely control giant vesicles containing magnetic microparticles
via externally operated magnetic fields. The monitored actuations established laminar flow patterns
suitable for the precise advection of the hybrid capsules at low Reynolds number, opening new
possibilities for the on-command transport of minute quantities of drugs, fluids or microobjects. The
encapsulation of magnetic particles also facilitates the release of the contained substances, based on
near infrared laser pulse-triggered destabilization of the phospholipid membranes.
We are also interested in the transport of microsized particles on confined environments. Particles
adsorbed on fluid interfaces or in the proximity of solid surface, present an interesting behavior, since
this is the common situation in most of the real-life systems. The boundary conditions determine the
response of the system, which presents new states and new locomotion strategies.
Vesicles containing magnetic nanowires combine the transport at will of microscopic systems, the protection
conferred by the phospholipid membrane, and the required flexibility to deform during passage through
microscale openings

VESÍCULAS GIGANTES COMO NADADORES A BAJO NÚMERO DE REYNOLDS
RESUMEN
Algunos microorganismos reptan o nadan en ausencia de apéndices externos gracias al movimiento
coordinado de orgánulos internos a la membrana, que empujan a las células gracias a fuerzas
intracelulares. En nuestro grupo, hemos construido un nadador sintético en el que vesículas lipídicas
gigantes son transportadas y dirigidas a distancia mediante la aplicación de campos magnéticos
externos. Las actuaciones monitorizadas generan flujos laminares en el medio acuoso adecuados para
la advección precisa de los sistemas a bajo número de Reynolds, abriendo la posibilidad de introducir
nuevas estrategias en el transporte de fármacos, fluidos o micro-objetos. La encapsulación de los
componentes magnéticos también facilita la liberación de las sustancias transportadas, gracias a la
desestabilización de las membranas lipídicas a través de láseres pulsados de infrarrojo.
También estamos interesados en el transporte de partículas microscópicas en ambientes confinados.
Las partículas adsorbidas en interfases fluidas, o próximas a una interfase sólida, presentan un
comportamiento interesante, ya que esta es la situación más común en la mayoría de los sistemas
reales. Las condiciones de contorno determinan la respuesta del Sistema, que puede presentar nuevos
estados y nuevas vías de locomoción.

MEMBRANE ADHESION: FROM MITOCHONDRIAL DYNAMICS TO PHARMACEUTICAL
IMPLICATIONS
ABSTRACT
Mitochondria form a dynamic network of organelles that constantly fuse and divide to maintain the energy requirement of the cell. The optimal balance of the mitochondrial dynamics between these antagonistic processes is crucial for the normal functioning of the cell and strictly controlled by mitochondrial membrane proteins, mitofusin 1 (Mfn1) and mitofusin 2 (Mfn2). Together with Mfn2, Mfn1 is directly involved in the fusion of the mitochondrial outer membrane, though the underlying molecular mechanism is not well understood (1). The membrane fusion initiates with the docking and adhesion of opposing membranes leading to maturation and final fusion. The adhesion of mitochondrial membranes can be considered as a novel therapeutic strategy in cancer therapy. The mitochondria of cancer cells have unique hallmarks that differ from their healthy counterparts. An important characteristic of mitochondria of cancer cells that differ from their healthy counterparts is the presence of a stronger membrane potential, which can be exploited to specifically accumulate cytotoxic cationic drugs designed for therapy. Here we explore the cytotoxic effect the lipophilic and positively charged molecule 10-N-nonyl acridine orange (NAO) that promotes strong membrane adhesion of negatively charged lipid membranes (2, 3). The attractive forces are derived from van der Waals interactions between anti-parallel H-dimers of NAO molecules from opposing bilayers. This strong adhesion and membrane remodeling interfere with mitochondrial dynamics leading to apoptosis when incubated at high micromolar concentration. As a proof-of-concept we treated H520 cancer cells with NAO as potential therapeutic drug and found as predicted an increased cytotoxicity (up to 2 orders of magnitude) when compared to healthy control (4).

ADHESIÓN DE MEMBRANA: DESDE LA DINÁMICA MITOCONDRIAL A SUS
IMPLICACIONES FARMACÉUTICAS
RESUMEN
Las mitocondrias forman una red dinámica de orgánulos que se fusionan y dividen constantemente
para mantener la energía necesitado por la célula. El balance óptimo de la dinámica mitocondrial entre
estos procesos antagonistas es crucial para el funcionamiento normal de la célula y están
estrictamente controlados por las proteínas de membrana, mitofusina 1 (Mfn1) y mitofusina 2 (Mfn2)
responsables para promover la fusión de la membrana externa mitocondrial, aunque el mecanismo
molecular se desconoce. La fusión de la membrana se inicia con el acoplamiento y la adhesión de
membranas mitocondriales adyacentes que a continuación conducen a la maduración y finalmente a
la fusión. El conocimiento detallado de la adhesión de las membranas mitocondriales puede
considerarse como una nueva posible diana para el diseño de terapia oncológica. Las mitocondrias de
las células cancerosas poseen, diferentes a las presentes en células sanas, un mayor potencial de
membrana que puede ser usado para acumular específicamente moléculas catiónicas citotóxicas. El
colorante fluorescente naranja 10-N-nonil acridina (NAO), una molécula lipofílica catiónica, es usado
generalmente como marcador mitocondrial con efectos citotóxicos en cultivos células eucariotas
cuando se incuban con altas concentraciones (µM) que promueve la adhesión de las membranas
mitocondriales induciendo la muerte celular o apoptosis. Utilizando técnicas ópticas, que incluyen
microscopía confocal de fluorescencia y microscopía de imagen de tiempos de vida media (FLIM) tanto
en fibroblastos embrionarios de ratón como en membranas modelo usando vesículas gigantes
unilamelares (GUVs) en combinación con simulaciones mediante dinámica molecular revelamos
mecanismo molecular subyacente al efecto citotóxico del NAO en células sanas y células H520 de
carcinoma de pulmón humano. Esta prueba de concepto donde observamos una mayor citotoxicidad
del NAO en las células cancerosas H520 en comparación con células sanas permite su uso como un
potencial fármaco terapéutico el tratamiento oncológico.

HIGH PRESSURE AS A STRATEGY IN THE DESIGN AND CHARACTERIZATION OF
MATERIALS
ABSTRACT
Pressure, as a physical variable, is defined by the amount of force acting per unit area. It is worth
mentioning that pressure, as thermodynamic variable, has the widest range of variation, from the 10-
32 bar existing in the interstellar space to the 1032 bar at which some processes take place inside some
stars. Pressure can be used for synthesizing new materials (by combining high pressure and high
temperature conditions) and for characterizing materials (optical, magnetic, and transport properties).
Our work in focused on studying different kind of materials susceptible to change their properties
when external pressure is applied on. We use different techniques for studying samples under the high
pressure conditions: experimental and theory. We have different kind of high pressure cells, three
different Raman Spectrometers, XRD diffractometer, resistivity measurements setup and knowledge
for DFT calculations.
In one hand, we are interested on Transition Metal Dichalcogenides (TMDs) with MX2 stoichiometry.
They are layered materials formed by stacking of 2D layers, commonly linked through weak Van der
Waals interlayer interactions. TMDs have some properties similar to graphite or even few-layers
graphene. They therefore have become interesting potential technological materials for applications
in the electronics and optoelectronics fields. The exotic phenomena found in these systems can be
tuned by both temperature and pressure. We are particularly interested on HfSe2 due to its exhibited
attractive electronic properties (semiconductor with a band gap close to 1 eV and high electronic
mobility).
On the other hand, transition metal oxides are attracting a growing interest due to their useful
properties for different applications. Among these oxides, the perovskite structure can accommodate
most of the metallic cations of the Periodic Table, which define their physical and chemical properties
and thus their technological applications. We have recently studied RCu3Mn4O12 (R= rare earth)
perovskite family. We can tune their magnetotransport properties by combining different metallic
cations in both A and B sites of the structure. We have used high pressure not only for the synthesis
but also for studying the behavior of these oxides under pressure. As a result, properties can be tuned
by chemical internal pressure and by application of external pressure due to the change in the cation
interactions.

RESUMEN
La Presión, como variable física, se define como la fuerza aplicada por unidad de área. Merece la pena
destacar, que la presión como variable termodinámica, tiene el mayor rango de variación, desde 10-32
bar que encontramos en el espacio interestelar hasta los 1032 bar a lo que ocurren algunos sucesos en
el interior de estrellas de neutrones. La presión se puede usar tanto para la síntesis de nuevos
materiales (combinando condiciones de alta presión y alta temperatura) como para la caracterización
de materiales (desde el punto de vista óptico, magnético, electrónico y de propiedades de transporte).
El trabajo de nuestro grupo de investigación está enfocado en el estudio de diferentes materiales que
son susceptibles de modificar sus propiedades cuando se les aplica una presión externa. Utilizamos
diferentes técnicas de caracterización bajo condiciones de alta presión: tanto experimentales como
cálculos teóricos. Entre otros, disponemos en nuestro grupo de diferentes celdas de alta presión,
equipos de espectroscopía Raman, difractómetro de rayos X, sistema de medidas de transporte y
conocimientos necesarios para llevar a cabo cálculos DFT.
A modo de ejemplo, por un lado, estamos interesados en los Dicalcogenuros de Metales de Transición
(TMDs) con estequiometría MX2. Estos son materiales laminares formados por el apilamiento de
láminas bidimensionales, que están unidas entre sí a través de interacciones débiles de Van der Waals.
Los TMDs poseen propiedades similares a las de grafito o incluso a las de grafeno de pocas capas,
convirtiéndose en materiales de alto interés con potencial tecnológico en el campo de electrónica y
optoelectrónica. La ventaja de estos materiales sobre el grafeno es que sus propiedades son intrínsecas
y no tienen que ser funcionalizadas como en el caso del grafeno. La fenomenología exótica que
presentan los TMDs se puede sintonizar a través de cambios en la temperatura o en la presión. Ahora
estamos particularmente interesados en el HfSe2 debido a que exhibe unas propiedades electrónicas
muy atractivas: semiconductor con un gap pequeño de alrededor de 1 eV y con una alta movilidad
electrónica.
Por otro lado, los óxidos de metales de transición han tenido un interés creciente en los últimos años
debido a sus propiedades adecuadas para diferentes tipos de aplicaciones. Entre estos óxidos, la
estructura perovskita y sus derivados constituyen una de las familias de materiales más fructíferas en
los campos de Materia Condensada y Ciencia de Materiales. Esta estructura incluye un amplio rango
de compuestos ya que puede acomodar en su estructura a la mayoría de cationes metálicos de la Tabla
Periódica, lo que define sus propiedades físicas y químicas y por lo tanto sus posibles aplicaciones
tecnológicas. Recientemente hemos estudiado la familia de perovskitas RCu3Mn4O12 (R= tierra rara).
Podemos sintonizar sus propiedades de magnetotransporte combinando diferentes cationes metálicos
en sus posiciones A y B de la estructura. Hemos utilizado la alta presión no sólo para su síntesis sino
también para estudiar el comportamiento de estos óxidos bajo presión. Como resultado, las
propiedades se pueden sintonizar mediante la presión química y por la aplicación de una presión
externa debido al cambio de las interacciones entre cationes.

DEVELOPMENT OF MATERIALS FOR ELECTROCHEMICAL ENERGY STORAGE SYSTEMS
ABSTRACT
Energy storage systems, such as supercapacitors and batteries, have attracted much interest in the last
decades due to the increase in energy consumption, together with the depletion of conventional
resources, based on fossil fuels, and the impact that their overuse has on climate change. Efforts have
been focused especially on the development of electrochemical energy storage systems that should
meet market requirements such as low cost, efficiency and safety. In the particular case of
supercapacitors, many materials such as porous carbons, transition metal oxides or conducting
polymers have been tested as electrodes. Pseudocapacitive transition metal oxide nanoparticles, such
as NiO or manganese oxides (MnOx) are promising candidates as electrodes due to their high specific
capacitance, low cost, abundance, safety and environmentally benign nature. Nonetheless, these
excellent properties are limited by their poor electrical conductivity, cycling stability and rate
capability. On the contrary, although carbon-based materials exhibit superior electrical conductivity,
long cycle life and high-power density, their lower specific capacitance is unsatisfactory for practical
applications where high energy density is required. Consequently, in this communication our
investigations devoted on the development of new electrode architectures based on the combinations
of the previous materials are described.

DESARROLLO DE MATERIALES PARA SISTEMAS DE ALMACENAMIENTO DE ENERGÍA
ELECTROQUÍMICA
RESUMEN
Los sistemas de almacenamiento de energía, como los supercondensadores y las baterías, han
suscitado un gran interés en las últimas décadas debido al aumento exponencial en el consumo de
energía, unido al agotamiento de los recursos convencionales, basados en combustibles fósiles, y el
impacto que su uso excesivo tiene sobre el cambio climático. Los esfuerzos se han centrado en el
desarrollo de sistemas de almacenamiento de energía electroquímica que deben cumplir los requisitos
del mercado, como el bajo coste, eficiencia y seguridad. En el caso particular de los
supercondensadores, muchos materiales como los carbonos porosos, los óxidos de metales de
transición o los polímeros conductores han sido utilizados como electrodos. Las nanopartículas de
óxidos de metales de transición pseudocapacitivas, como el NiO o los óxidos de manganeso (MnOx)
son candidatos prometedores como electrodos debido a su alta capacidad específica, bajo coste,
abundancia, seguridad y bondad medioambiental. No obstante, estas excelentes propiedades están
limitadas por su baja conductividad eléctrica, pobre estabilidad de ciclado y una importante
disminución de la capacidad con la velocidad de ciclado. Por el contrario, aunque los materiales
basados en carbono exhiben una conductividad eléctrica superior, una vida útil prolongada y una
densidad potencia mayor, su baja capacidad específica no resulta satisfactoria para aplicaciones
prácticas donde se requiere una alta densidad de energía. En consecuencia, en esta comunicación se
describen nuestras investigaciones dedicadas al desarrollo de nuevas arquitecturas de electrodos
basadas en las combinaciones de los materiales anteriores.

NOVEL MATERIALS FOR MORE SUSTAINABLE ELECTROCHEMICAL ENERGY STORAGE
ABSTRACT
In a worldwide increasing population, which consumes an increasing amount of energy, fully
benefitting from renewables requires energy Storage. Electrochemical energy storage in batteries and
capacitors, is spreading due to their modularity and flexibility. For large scale applications, more
sustainable electrochemical energy storage devices are seeked via one or several of the following
routes: i) more abundant alkali metal ions as driving vectors (Sodium or Potassium ion batteries rather
than Lithium based devices; ii) the use of more abundant and easy to extract raw materials (such as
those based on abundant metals like iron or organic compounds); iii) with materials produced via
sustainable routes such as Prussian Blue based cathodes or Schiff bases based anodes (polymers, salts);
iv).directly processable multifunctional materials such as block co-polymers. The objective of the
research line is not only to find sustainable materials with good energy storage performance, but also
to understand their mechanism of electrochemical reaction and failure during charge storage by
means of ex-situ as well as in-situ techniques such as spectroscopy, powder X-ray diffraction or
transmission electron microscopy.

EN LA BÚSQUEDA DE NUEVOS MATERIALES PARA UN ALMACENAMIENTO
ELECTROQUÍMICO DE ENERGÍA MÁS SOSTENIBLE
RESUMEN
En una sociedad que consume cada vez mayores cantidades de energía, la transición hacia un mayor
aprovechamiento de energías renovables requiere el uso de almacenamiento de energía,
extendiéndose cada vez más el almacenamiento electroquímico en baterías ya que son modulares y
muy flexibles. En aplicaciones de gran escala, es necesario buscar mejoras que hagan estos dispositivos
más sostenibles por algunas o varias de las siguientes rutas: i) el uso de dispositivos basados en metales
más abundantes (baterías de ión sodio o potasio frente a las baterías de ión litio); ii) empleando
materiales cuyas materias primas sean abundantes y fáciles de extraer (materiales basados en
compuestos orgánicos y metales abundantes como el hierro); iii) que se puedan sintetizar por rutas
más sostenibles, como los cátodos basados en azules de Prusia o como los ánodos basados en bases
de Schiff (polímeros, sales, COFs); iv) materiales multifuncionales que puedan procesarse directamente
tales como los copolímeros de bloque. El objetivo no sólo es encontrar materiales con buenas
prestaciones que cumplan con estos criterios de sostenibilidad, sino también estudiar los mecanismos
de operación durante su ciclado electroquímico por técnicas de difracción de rayos X o microscopía
electrónica de transmisión, in-situ durante el ciclado electroquímico para entender los mecanismos de
degradación y falla. Uno de los aspectos clave en adelante será encontrar y estudiar materiales que
puedan operar como ánodos en baterías de ión potasio.

THE ROLE OF CHEMISTRY IN THE FUNCTIONALITY OF MAGNETORESISTANT
MATERIALS
ABSTRACT
Looking for new ways to improve material properties constitutes an intensive research area for
technological development. The perovskite structure has traditionally attracted a great interest since
properties such as colossal magnetoresistance has been described in materials showing related
structures. Moved by the obvious technological interest of that property this project is focussed on the
design, synthesis and characterization of new manganese oxides related to perovskite structure. The
introduction of both cationic and anionic vacancies seems to be the key to tailor advanced functional
materials with high values of the colossal magnetoresistance above room temperature. Aberration-
corrected scanning transmission electron microscopy technique combined with electron energy loss
spectroscopy is crucial to the understanding of the nanophase separation and consequently, of the
electrical and magnetic behavior of these perovskites.
Figure shows the temperature dependence of the magnetoresistance for La0.5Ca0.5MnO3 samples with different
% of cationic vacancies.

RESUMEN
La búsqueda de nuevas vías para mejorar las propiedades de los materiales constituye un área de
investigación intensiva para el desarrollo tecnológico. Materiales derivados del tipo estructural
perovskita han atraído tradicionalmente un gran interés ya que propiedades como la
magnetorresistencia colosal se han descrito en materiales que muestran estructuras relacionadas.
Movidos por el evidente interés tecnológico de esa propiedad, este proyecto se centra en el diseño,
síntesis y caracterización de nuevos óxidos de manganeso relacionados con la estructura perovskita.
La introducción de vacantes catiónicas y aniónicas parece ser la clave para conseguir materiales
funcionales avanzados con altos valores de magnetorresistencia colosal por encima de temperatura
ambiente. La microscopía electrónica con resolución atómica, combinada con técnicas
espectroscópicas asociadas, ha permitido proponer un modelo que explica el origen de la separación
de fases magnéticas y eléctricas en estos sistemas.

SYNTHESIS, CHARACTERIZATION AND PROPERTIES OF RUTHENIUM DINUCLEAR
COMPOUNDS WITH METAL-METAL BOND
ABSTRACT
Diruthenium compounds with metal-metal bond and paddlewheel structure can be obtained by means
of several synthetic methods like refluxing conventional synthesis, solvothermal synthesis or
microwave assisted solvothermal synthesis. The magnetic properties of the Ru(II)-Ru(III) compounds
are of special interest among their properties, since they do not only have three unpaired electrons,
being 3/2 spin systems, but they also show a strong zero field splitting and antiferromagnetic
interactions between the dimetallic Ru25+ units. On the other hand, some lanthanides ions have a high
number of unpaired electrons, large magnetic anisotropy, and luminescence. In this research line, the
obtained 2D and 3D structures by the combination of [Ru2(CO3)4]3- and lanthanide ions are studied in
order to get multifunctional materials with interesting magnetic and luminescence properties.
Furthermore, the open-paddlewheel compound [Ru2Cl2(μ-DPhF)3(DMSO)] (DPhF = N,N’-
diphenylformamidinate) has been observed to interact with the RNA molecule, preferentially attacking
to nucleotides situated at or close to the junctions of the RNA structure. This fact has opened a very
interesting research line in which the kind of interaction of ruthenium dinuclear complexes with
nucleotides and other biological systems are been studied.

RESUMEN
Los compuestos de dirrutenio con enlace metal-metal y estructura paddelwheel pueden obtenerse
mediante diversos métodos sintéticos, como la síntesis convencional a reflujo, la síntesis solvotermal
o la síntesis solvotermal asistida por microondas. Entre las propiedades de los compuestos de Ru(II)-
Ru(III) destacan sus propiedades magnéticas, ya que, no solo tienen tres electrones desapareados,
comportándose como sistemas de espín 3/2, sino que también presentan un fuerte desdoblamiento a
campo cero e interacciones antiferromagnéticas entre las unidades dimetálicas Ru25+. Por otro lado,
algunos iones lantánidos presentan un elevado número de electrones desapareados, alta anisotropía
magnética y luminiscenia. En esta línea de investigación se estudian las estructuras 2D y 3D obtenidas
tras la unión de [Ru2(CO3)4]3- con iones lantánidos para la obtención de materiales multifucionales con
propiedades magnéticas y luminiscentes interesantes.
Por otro lado, se ha observado que el compuesto con estructura open-paddlewheel Ru2Cl2(μ-
DPhF)3(DMSO)] (DPhF = N,N’-difenilformamidinato) interacciona con el ARN atacando
preferentemente a nucleótidos situados en las intersecciones, o próximos a ellas, de la estructura del
ARN. Esto ha abierto una línea de investigación muy interesante en la que se está estudiando el tipo
de interacción de complejos dinucleares de rutenio con nucleótidos y con otras especies biológicas.

MOLECULAR ASSEMBLIES BASED ON COORDINATION COMPOUNDS
ABSTRACT
The main aim of this research is the synthesis, characterization and study of coordination compounds
with Schiff base ligands based on bicompartmental “salen” type. These kind of ligands have two
cavities of different nature and size: an internal N2O2 suitable for 3d (MT), and an external O2O2 one
able to host alkaline, alkaline-earth, 3d transition metals ions, 4f metal ions and small molecules such
as water, ammonium or hydronium ions which can form hydrogen bonds. The Interaction of the metal
ions with the two cavities will be different depending on their nature and size, resulting in a wide
structural variety, with different stoichiometries and topologies, which gives rise to different
supramolecular networks. The structural characteristics are closely related to their properties
(electronic, magnetic, electrical and luminescent...), as well as their potentials applications. For that
reason, a deep structural study of the complexes by single X-ray crystallography is essential.
In that sense, this type of compounds shows a great variety of applications in different fields.
Therefore, different types of complexes, containing diverse bicompartmental ligands, have been
designed and studied depending on the desired properties and the target applications. Concretely, the
studied systems are: i) MT(Cu, Ni)/ns block complexes, as structural models of biological ionophores;
(ii) MT(Cu, Ni)/ns block /TCNQ ligand systems, as building blocks to obtain compounds with magnetic
and/or electrical properties; (iii) MT(Cu, Ni)/MT for obtaining luminescent materials; (iv) Cu/Cu and
Cu/Mn systems as structural/functional models of catecholases.

ENSAMBLAJES MOLECULARES BASADOS EN COMPUESTOS DE COORDINACIÓN
RESUMEN
El objetivo fundamental de nuestra investigación es la síntesis, caracterización y estudio de complejos
de coordinación con bases de Schiff basadas en ligandos bicompartimentales tipo “salen”. Estos
ligandos bicompartimentales presentan dos cavidades de diferente naturaleza y tamaño: una interna
N2O2 adecuada para alojar iones metálicos 3d (MT) y otra externa O2O2 capaz de alojar iones alcalinos,
alcalinotérreos, metales de transición 3d, iones 4f y pequeñas moléculas como agua, iones amonio u
oxonio que pueden formar enlaces de hidrógeno. Las interacciones de los iones metálicos con las
distintas cavidades serán diferentes en función de su naturaleza y tamaño, obteniéndose una amplia
variedad estructural, con diversas estequiometrías y topologías, dando lugar a diferentes redes
supramoleculares. Las características estructurales están íntimamente relacionadas con sus
propiedades (electrónicas, magnéticas, eléctricas o luminiscentes…), así como con sus potenciales
aplicaciones. Por ello, es fundamental un detallado estudio estructural mediante difracción de rayos X
de monocristal.
En este sentido, este tipo de compuestos presentan muy variadas aplicaciones en diferentes campos.
Por ello, se han diseñado y estudiado diferentes tipos de complejos, conteniendo distintos ligandos
bicompartimentales, en función de las propiedades y aplicaciones buscadas. Concretamente, se han
estudiado los sistemas: i) MT(Cu, Ni)/bloque ns, como modelos estructurales de ionóforos biológicos
entre otras posibles aplicaciones; ii) MT(Cu, Ni)/bloque ns/ligando TCNQ como bloques de
construcción para obtención de compuestos con propiedades magnéticas y/o eléctricas; iii) MT(Cu,
Ni)/MT para la obtención de materiales luminiscentes; iv) sistemas Cu/Cu y Cu/Mn como modelos
estructurales/funcionales de catecolasas.

BIOMOLECULE AS SCAFFOLD TO CREATE HIGHLY ORDERED n/p-MATERIALS
ABSTRACT
The control on the organization and morphology of organic materials at different scales is an essential
challenge in current science. In particular, organic materials employed for obtaining efficient
photovoltaic devices require a controlled segregation of electron donor/acceptor domains in the active
layers because transport of the photogenerated charge carriers occurs through these domains to the
electrodes. This control over the organization of nanostructured domains at the same length scale
generally results in an increase in conductivity or photoconductivity values. One of the approaches to
prepare optoelectronic materials for photon-energy conversion is the use of covalent donor−acceptor
(D-A) dyads. However, the precise control over the organization of photoactive components is still a
challenge.
With this goal in mind, our research focuses on the use of biomolecule such as peptide and protein to
organize photo- and electroactive units to achieve highly ordered functional materials. Initially, an
electron donor π-extended tetrathiafulvalene and an electron-acceptor perylene-bisimide were self-
organized separately obtaining n and p-nanofibers at the same scale. These complementary n and p-
nanofibers are endowed with ionic groups with opposite charges on their surfaces. The synergic
interactions establish periodic alignments between both nanofibers resulting in a material with
alternately segregated donor/acceptor nanodomains with photocurrent value up to 0.8 cm2 V−1 s−1,
confirming the effectiveness in the design of these heterojunction structures.
Another approach is the use of a designed repeat protein scaffold with unique features that allow the
control of their structure, functionality, and assembly. This design leads to the organization of
porphyrin molecules and also the formation of n/p-hybrids materials wrapping SWCNT with
photocurrent values.

RESUMEN
El control de la organización y morfología de los materiales orgánicos a diferentes escalas es un reto
esencial en la ciencia actual. En particular, los materiales orgánicos empleados para obtener
dispositivos fotovoltaicos eficientes requieren una segregación controlada de los dominios
dadores/aceptores de electrones en las capas activas, ya que el transporte de los portadores de carga
fotogenerados se produce a través de estos dominios hasta los electrodos. Este control sobre la
organización de dominios nanoestructurados, a diferentes escalas de longitud, generalmente resulta
en un aumento de los valores de conductividad o fotoconductividad. Uno de los enfoques para
preparar materiales optoelectrónicos para la conversión de energía fotónica es el uso de díadas de
dadores-aceptor (D-A). Sin embargo, el control preciso sobre la organización de los componentes
fotoactivos sigue siendo un reto.
Con este objetivo en mente, nuestra investigación está dirigida al uso de biomoléculas tales como
péptidos y proteínas, como plataformas para la organización de unidades foto y electroactivas con el
fin de lograr materiales funcionales altamente ordenados. Inicialmente, nuestro primer estudio se
basó en el uso de la unidad dadora de electrones, tetrathiafulvalene π-extendido, covalentemente
unido a una secuencia peptídica y unidad aceptora de electrons perilenbiimida. Esta secuencia
péptidica permite la formación de láminas y con ello la obtención de nanofibras. La capacidad de
ambas moléculas para auto-asociarse permite la formación instantánea del material co-ensamblando.
Las propiedades más interesantes de dichos materiales se hallaron mediante estudios de microscopía
de rayos-X, mostrando estructuras fibrilares organizados desde la nano hasta la macroescala
compuesta por dominios segregados de dadores y aceptores. Además, estos materiales mostraron
valores de fotocorriente en torno a 0.8 cm2 V-1 s-1, lo que confirma la eficacia en el diseño de estas
estructuras de heterounión.
Otra aproximación que hemos llevado a cabo es el uso de proteínas TPR como andamio para la
organización tanto en disolución como en estado sólido. Esta aproximación nos ha permitido un
preciso control de la organización de las unidades electroacitivas, así como la obtención de materiales
híbridos dador-aceptor con buenos valores de fotocorriente.

NMR AND MOLECULAR RECOGNITION
ABSTRACT
We are working on the study of molecular recognition events by using NMR. Our research lines are
focus on the characterization of complex carbohydrates (mainly N-glycans) and their interactions. In
addition, we are developing new NMR methodologies that allow studying those biomolecules that
could not be characterize by standard methods due to their pseudo-symmetry or molecular size.
Complex glycosylation patterns containing multiantennary N-glycans are typically found in human
glycoproteins. However, the structural characterization of these glycans is rather challenging. Usually,
standard NMR and X-ray diffraction techniques fail to provide specific answers on the structure and
molecular recognition features due to intrinsic attributes of the glycan. As a promising approach,
carbohydrates conjugated to lanthanide binding tags have revealed high potential toward this aim.
This methodology has first been applied to the study of small oligosaccharides (di-, tri- and
tetrasaccharides). Proceeding from this experimental basis, we have extended this concept to the level
of high degree branching and long chain N-glycans. In addition, we are able to study the molecular
recognition properties of these complex glycans thanks to the unprecedented resolution obtained in
the spectra.

RESUMEN
Nuestra investigación se centra en el estudio de procesos de reconocimiento molecular empleando
Resonancia Magnética Nuclear, principalmente caracterizando carbohidratos complejos (N-glicanos) y
sus interacciones. Asimismo, estamos trabajando en el desarrollo de nuevas metodologías que
permitan aplicar la RMN a aquellas biomoléculas que por su pseudo-simetría y tamaño continúan
siendo un reto en el ámbito de la biología estructural.
Las glicoproteínas humanas presentan habitualmente patrones de glicosilación complejos. Sin
embargo, la caracterización estructural de estos carbohidratos continúa siendo un reto. Normalmente,
los métodos de RMN standard y difracción de rayos-X no permiten caracterizar en detalle estas
moléculas debido a sus peculiaridades estructurales. En este contexto, los carbohidratos conjugados
con agentes quelantes de lantánidos permiten solventar muchos de los inconvenientes de los métodos
tradicionales. Esta metodología se aplicó inicialmente al estudio de oligosacáridos pequeños (di-, tri- y
tetrasacáridos) y posteriormente hemos extendido el método al estudio de carbohidratos
multiantenarios y de cadena larga. El incremento espectacular que obtenemos en la resolución
espectral nos ha permitido además de caracterizar la conformación de las moléculas obtener los
determinantes estructurales que gobiernan el reconocimiento molecular de estos oligosacáridos.

STRATEGIES AND MOLECULAR RECOGNITION
ABSTRACT
The development of chemical sensors and molecular probes is a multidisciplinar research with
applications in many diverse fields: from environmental monitoring, food safety and biomedical and
pharmacological appilcations, to the control of industrial processes or personal protection. In this
respect, having selective, sensitive and rapid ways of detecting the species of interest is of vital
importance. Fluorescence (or more generally, luminescence)-based detection methods offer superb
sensitivity, multiparametric information and time and spatial resolution, with a straightforward
handling, being cheaper than other well-stablished analysis techniques such as chromatography. In
our research group we are focused on the development of new optical chemosensors and fluorescent
probes being involved in the whole discovery process: from luminophore design and synthesis,
photochemical and photophysical characterization (with the elucidation of signalling mechanisms for
the modulation of the photoinduced processes involved in chemosensor response), to the final
analytical application. Selectivity in the final sensor or assay is implemented by making use of
supramolecular concepts for ligand design, use of biological recognition elements, or use of biomimetic
recognition materials such as molecularly imprinted polymers (MIPs). Concepts of (nano)materials
chemistry also helps in sensor design, and offers the possibility of a modular design (i.e. core-shell
nanoparticles) and improvement of the recognition and signaling properties (signal amplification,
improved guest binding by modulating the chemical properties of the materials, introducing shape and
size selectivity by the creation of specific binding cavities such as in MIPs, etc). Our main goals are food
safety control and medical or biochemical monitoring. Instrumental facilities in our laboratories
include UV-Vis spectrophotometers, several spectrofluorometers for steady-state and time-resoved
measurements, a fluorescence lifetime imaging confocal microscope (FLIM), a laser kinetic
spectrometer with NIR detection, a microtiter plate reader with different reading modes and sensor
technology coupled to fiber optics.

RESUMEN
El desarrollo de sensores químicos y sondas moleculares es un área de investigación multidisciplinar
con aplicaciones en campos muy diversos: desde la monitorización medioambiental, seguridad
alimentaria y aplicaciones biomédicas y farmacológicas, al control de procesos industriales y la
protección personal. A este respecto, es de vital importancia poder disponer de medios de detección,
selectivos, sensibles y rápidos, de las especies de interés. Los métodos de detección de fluorescencia
(o más generalmente, de luminiscencia) ofrecen una sensibilidad excelente, información
multiparamétrica y resolución espacial y temporal. Su manejo es sencillo, siendo normalmente
métodos de análisis más económicos que otros bien establecidos como la cromatografía. En nuestro
grupo de investigación nos centramos en el desarrollo de nuevos sensores químicos ópticos y sondas
fluorescentes, estando implicados en todo el proceso: desde el diseño de los luminóforos y su síntesis,
la caracterización fotofísica y fotoquímica (con la elucidación de los mecanismos de señalización para
la modulación de los procesos fotoinducidos implicados en la respuesta del sensor químico), a la
aplicación analítica final. La selectividad en el sensor o bioensayo se implementa mediante el uso de
conceptos de Química Supramolecular para el diseño de ligandos, el empleo de elementos de
reconocimiento biológicos, o el uso de materiales biomiméticos de reconocimeinto como los
polímeros de impronta molecular (MIPs). Conceptos de química de (nano)materiales también
contribuyen en el diseño de los sensores, ofreciendo la posibilidad de un diseño modular (i.e. con
nanopartículas núcleo-recubrimiento) y mejora de las propiedades de reconocimeinto y señalización
(amplificación de la señal, mejora del enlace con el analito mediante la modulación de las propiedades
químicas de los materiales, introducción de selectividad geométrica mediante la creación de cavidades
específicas como en el caso de los MIPs, etc). Nuestros principales objetivos son el control en seguridad
alimentaria y la monitorización médica o bioquímica. La instrumentación disponible en nuestros
laboratorios incluye espectrofotómetros UV-Vis, espectrofluorímetros para medidas en el estado
estacionario y resueltas en el tiempo, un microscopio confocal para imagen por tiempo de vida de
fluorescencia (FLIM), un espectrómetro de cinética láser con detección NIR, un lector de placas de
pocillos con distintos modos de medida, y tecnología de sensores acoplada a fibra óptica.

NOVEL REACTIVITY OF ALLENES VIA METAL CATALYSIS
ABSTRACT
The research work carried out in my research group is intended to contribute to the development of
new methodologies based on cyclization and/or transposition reactions of allenes and cumulenes for
the preparation of structurally novel polycyclic systems, including molecules with potential biological
activity. The main conclusions of this study are summarized below:
1. The gold-catalyzed carbocyclization reactions of different allenes linked to biological importance
nucleus, such as the β-lactam ring and glucofuran sugar, have been investigated. The preparation of
medium-sized ring polycycles through selective C–H bond activation is a big challenge. In this regard,
the gold catalysed intramolecular hydroarylation of allenes may be a possible solution to produce
eight- or nine-membered carbocycles, although this achievement has not yet been accomplished. We
present here an unprecedented Au-catalyzed 9-endo carbocyclization of aryl allenes as a powerful
synthetic tool to obtain novel nine-membered annulated β-lactam derivatives. In addition, it is shown
that the outcome of the reaction (9-endo hydroarylation versus formal 5-exo hydroalkylation) can be
modulated by the allene tether.
2. We present a novel and unanticipated reactivity in gold catalysis starting from 3-allenyl 4-
aryl(alkenyl) lactams to lead strained adducts which show cage-like structures in a
diastereoselective manner.

RESUMEN
El trabajo de investigación que se lleva a cabo en mi grupo de trabajo pretende contribuir al desarrollo
de nuevas metodologías de ciclación y transposición de alenos y cumulenos catalizadas por metales
para la preparación de sistemas policíclicos estructuralmente novedosos, así como moléculas con
potencial actividad biológica. Las principales conclusiones de este estudio se resumen a continuación:
1. Se ha investigado la reacción de carbociclación catalizada por oro de diferentes alenos unidos
a núcleos de importancia biológica, como son el anillo β-lactámico y el azúcar glucofuranosa. Mientras
que las alenil-β-lactamas sufren una hidroarilación 9-endo para dar lugar a benzociclos fusionados de
nueve miembros, los alenilazúcares conducen a ciclopentenos bicíclicos a través de una
hidroalquilación 5-exo poco común.
2. Se ha descrito una inesperada síntesis diastereoselectiva de compuestos tensionados “tipo
caja” a partir de alenil-β-lactamas diferentemente sustituidas utilizando catálisis de oro. El proceso
implica la ruptura simultánea de los enlaces N1‒C4 y C2‒C3 del anillo β-lactámico y la formación
simultánea de varios enlaces carbono‒carbono.

IDENTIFICATION AND VALIDATION OF THERAPEUTIC TARGETS
ABSTRACT
In the Medicinal Chemistry Laboratory at Complutense University our work is focused on two main
research interests: the development of new pharmacological agents and the identification of
therapeutic targets.
In the first approach we are carrying out several research lines on three fields of major relevance for
human health nowadays: cancer and CNS-associated diseases. In this regard, we are developing new
compounds with antitumor activity (inhibitors of isoprenylcysteine carboxyl methyltransferase -ICMT-
enzyme) and agents acting at the central nervous system (monoacylglycerol lipase enzyme, MAGL).
Additionally, we are working on lines of research aimed at the design and synthesis of ligands for type
1 of the lysophosphatidic acid receptor (LPA1) and allosteric modulators of GPCRs. In the last years we
have also expanded our research to the design and synthesis of new antibiotics (inhibitors of FtsZ and
antibody-drug conjugates as new antituberculosis agents) as well as to the rare diseases field working
on progeria.
In the second approach, we are implementing forward chemical genomic methodologies in order to
identify therapeutic targets and to find new molecular entities. In this part of the work, our objective
is to develop methodologies that generate de novo information about the physiological and/or
pathological state of the biological system under study. In particular, we are implementing phenotypic
approaches to identify therapeutic targets and to find new molecular entities both in mammalian cells
and in the human microbiota. To achieve this goal, we are combining medicinal chemistry, chemical
biology, metabolomics and proteomics, expecting that the integration of these methodologies will
provide us with a reliable and complete picture of the complexity of the biological system under study.
In this talk the most recent advances towards the identification and validation of new therapeutic
targets focused on cancer and on the rare disease progeria will be covered

RESUMEN
La investigación que realiza el laboratorio de Química Médica de la Universidad Complutense de
Madrid tiene dos objetivos principales: (i) el desarrollo de nuevas moléculas candidatas a fármacos y
(ii) la identificación de dianas terapéuticas.
El primer objetivo está siendo abordado en dos áreas de gran relevancia actual: cáncer y patologías
neurodegenerativas. En este sentido, estamos desarrollando nuevos compuestos con actividad
antitumoral (inhibidores de la enzima isoprenilcisteina carboxilmetiltransferasa, ICMT) y también
compuestos con actividad en el sistema nervioso central (como por ejemplo inhibidores de la lipasa de
monoacilglicéridos o MAGL). Adicionalmente, estamos trabajando en otras líneas dirigidas al diseño y
síntesis de nuevos ligandos del receptor de ácido lisofosfatídico LPA1 y de moduladores alostéricos de
receptores acoplados a proteínas G (GPCRs). Finalmente, en estos últimos años hemos iniciado
también el abordaje de enfermedades infecciosas mediante el desarrollo de nuevos antibióticos, tales
como inhibidores de la proteína FtsZ, y conjugados anticuerpo-fármaco como agentes
antituberculosos. Por último, hemos iniciado una línea de trabajo dirigida a la búsqueda de nuevas
estrategias farmacológicas para el tratamiento de una enfermedad rara, la progeria.
En la segunda aproximación, en estos momentos estamos implementando las estrategias de forward
chemical genomics más adecuadas para identificar nuevas dianas terapéuticas, así como para
encontrar nuevas entidades químicas. En esta parte del trabajo, nuestro objetivo es el desarrollo de
metodologías que puedan generar información de novo acerca del estado fisiológico o patológico del
sistema biológico objeto de estudio. Para lograr este objetivo, en nuestro grupo de investigación
empleamos química médica, biología, metabolómica y proteómica de tal modo que el uso combinado
de todo ellos nos permite finalmente obtener información acerca del sistema biológico objeto de
estudio.
Mostraré los avances más recientes que estamos aplicando en nuestro grupo de investigación para la
identificación y validación de nuevas dianas terapéuticas, con especial interés en el desarrollo de
nuevos agentes antitumorales, y también para el tratamiento de la enfermedad rara progeria.

ALLOSTERIC MODULATORS OF GPCRS: A NEW APPROACH TO DRUG DISCOVERY
ABSTRACT
In recent years, novel approaches have emerged for drug development targeting G protein-coupled
receptors (GPCRs), one of the most important classes of drug targets. Major therapeutic advantages
can be achieved with allosteric modulators (AMs) that do not bind the orthosteric site for the
endogenous ligand, but act in conjunction with it, enhancing (PAM) or attenuating (NAM) its affinity,
potency and/or efficacy.
In this context, in the Medicinal Chemistry Laboratory we are interested in the identification of AMs
for class A and B GPCRs, which may lead to new selective and efficient therapeutic agents to address
major unmet medical needs. In particular, in this communication we have presented a project aimed
at the development of AMs of the dopamine D1 receptor (D1R) as a novel strategy for the treatment
of Parkinson’s disease (PD). To start the project, we built a structure-based pharmacophore model to
identify structural motifs for modulation of the receptor. Next, a virtual screening of ZINC database
compounds using this pharmacophore model and subsequent docking yielded thirty-five virtual hits,
which were assessed in vitro for allosteric modulation of the receptor. Among them, a biphenyl
derivative was identified as hit and entered into a medicinal chemistry program that led to compound
UCM-01212, a potent and selective D1R PAM. UCM-01212 increases the endogenous dopamine
maximal effect in a dose-dependent manner and its allosteric binding site has been studied through
mutagenesis and docking experiments. Importantly, this compound influences motor coordination in
a mouse model of PD, which sets a proof of concept of the therapeutic interest of a D1R PAM as a
novel strategy to tackle the disease. In a next optimization step, derivative UCM-01306 displayed good
pharmacological and pharmacokinetic profiles. UCM-01306 is currently under evaluation in
collaboration with Kaertor Foundation and Johnson & Johnson to develop a drug candidate that could
validate the therapeutic utility of an AM of D1R for the treatment of PD.

RESUMEN
En los últimos años, han surgido nuevas aproximaciones para el desarrollo de fármacos dirigidos a los
receptores acoplados a proteínas G (GPCRs), unas de las dianas terapéuticas más importantes de la
industria farmacéutica. Entre estas estrategias, destaca el empleo de moduladores alostéricos
(allosteric modulators, AMs) debido a las ventajas terapéuticas que ofrece respecto a los ligandos
clásicos. Los AMs no se unen al sitio ortostérico del ligando endógeno para desplazarlo, sino que actúan
conjuntamente con él, mejorando (PAM) o atenuando (NAM) su afinidad, potencia y/o eficacia.
En este contexto, en el Laboratoria de Química Médica estamos interesados en la identificación de
AMs para GPCRs de las clases A y B que puedan conducir a nuevos agentes terapéuticos selectivos y
eficientes. En particular, en esta comunicación se presenta un proyecto dirigido al desarrollo de AMs
del receptor D1 de dopamina como una estrategia novedosa para el tratamiento de la enfermedad de
Parkinson (EP). El punto de partida del proyecto consistió en la elaboración de un modelo de
farmacóforo basado en la estructura del receptor que permitió identificar los motivos estructurales
claves para la modulación de su actividad. A continuación, un screening virtual de los compuestos
recogidos en la base de datos ZINC usando este modelo de farmacóforo, seguido de un análisis
mediante docking, nos proporcionó treinta y cinco candidatos que fueron evaluados in vitro. Entre
ellos, se identificó un derivado de bifenilo, que tras un proceso de optimización condujo a UCM-01212,
un nuevo AM del receptor D1 potente y selectivo. UCM-01212 aumenta el efecto máximo producido
por la dopamina endógena de manera dosis-dependiente, y su sitio de unión alostérico ha sido
estudiado mediante experimentos de mutagénesis y docking. La evaluación de UCM-01212 en un
modelo animal de la EP mostró que el compuesto es capaz de aumentar la coordinación motora, lo
que ha permitido establecer una prueba de concepto del interés de un PAM del receptor D1 como una
estrategia novedosa para abordar la enfermedad. En una etapa posterior de optimización, se obtuvo
el derivado UCM-01306, que posee buenos perfiles farmacológicos y farmacocinéticos. UCM-01306
está siendo evaluado actualmente en colaboración con la Fundación Kaertor y Johnson & Johnson con
el objetivo final de desarrollar un candidato a fármaco que permita validar la utilidad terapéutica de
un AM del receptor D1 para el tratamiento de la EP.

POSTRANSLATIONAL MODIFICATIONS: A NOVEL STRATEGY TO FIGHT METASTATIC
CANCER
ABSTRACT
Breast cancer is one of the most prevalent and the leading causes of mortality in the Western world.
The majority of breast cancer deaths result from the progression of this disease to a metastatic
phenotype. Preventing metastasis would make cancer a chronic, not fatal disease. Therefore,
improving our knowledge of the molecular and cellular mechanisms responsible of tumor cells
dissemination would facilitate the clinical management of this disease and, in turn, the identification
of new therapeutic targets, and the development of more effective treatments against metastatic
cancer. My groups is focused in the study of different regulators of cell mobility, such as SNAI2, NCAPH
or the GTPase RAC1. Previous work of the applicant group has shown that the SUMOylation of the
GTPasa RAC1 is required for the maintenance of the active form of RAC1 during the cell migration and
invasion. Furthermore, we have shown that RAC-SUMO is required for invasion in breast cancer cells
but not to other biological processes in which RAC1 participates, such as proliferation or survival. The
design of a peptide that interacts with the polybasic region of RAC1, where the lysines are likely to be
SUMOylated, has allowed us to raise the working hypothesis contemplated in this proposal: The
inhibition of RAC1 SUMOylation can be used to prevent the migration/invasion of tumor cells and,
therefore, metastasis.

MODIFICACIONES POSTRADUCCIONALES: UNA NUEVA ESTRATEGIA EN TERAPIA
CONTRA EL CÁNCER METASTÁSICO
RESUMEN
El cáncer de mama es uno de los más prevalentes y una de las principales causas de mortalidad en el
Mundo Occidental. La mayoría de las muertes por cáncer de mama son el resultado de la progresión
de esta enfermedad a un fenotipo metastásico. La prevención de metástasis convertiría al cáncer en
una enfermedad crónica, no mortal. Por tanto, mejorar nuestro conocimiento de los mecanismos
moleculares y celulares responsables de la diseminación de las células tumorales facilitaría el manejo
clínico de esta enfermedad y, a su vez, la identificación de nuevas dianas terapéuticas, así como el
desarrollo de tratamientos más efectivos contra el cáncer metastásico. Mi grupo se centran en el
estudio de diferentes reguladores de la movilidad celular, como SNAI2, NCAPH o la GTPasa RAC1.
Trabajo previo del grupo solicitante ha puesto de manifiesto que la SUMOilación del GTPasa RAC1 es

células de cáncer de mama, pero no para otros procesos biológicos en los que RAC1 participa, como la
proliferación o supervivencia. Hemos diseñado un péptido que interactúa con la región polibásica de
RAC1, donde se encuentran las lisinas que se SUMOilan, lo que ha permitido plantear una nueva línea
la hipótesis de trabajo contemplada en esta propuesta: La inhibición de la SUMOilación RAC1 se puede
utilizar para prevenir la migración / invasión de células tumorales y, por lo tanto, metástasis.

FUNCTION RELATIONSHIP OF THE PROTEINS OF PULMONARY SURFACTANT SP-B AND
SP-C
ABSTRACT
The lung surfactant is a lipid-protein complex which reduces the surface tensión at the alveolar air-liquid interface. The surfactant protein C (SP-C) is a highly conserved transmembrane protein of 35 amino acids. It’s involved in the transport of surfactant molecules in order to reduce the surface tension at the pulmonary interface. Due to its high hydrophobicity, in this project two systems of nanocomplexes that allow their solubilization are studied. These are membrane patches which can be bounded on the one hand by two copies of the scaffold protein MSP1D1 or on the other hand by the styrene-maleic acid copolymer SMA. Biophysical studies were performed on both nanodiscs including or not native porcine SP-C, or a recombinant human version (rSP-C). MSP1D1 nanodiscs have similar interfacial activity in the absence and presence of protein, and they inhibit the surfactant activity. SMA nanodiscs in the presence of protein seem to accelerate lipid adsorption at the interface, altering minimally the surfactant function. Although these nanodiscs seem to be more strongly anchored to phospholipid membranes than the empty nanodiscs, both are capable of being excluded from the interface, which is interesting for their possible use as a drug carrier into the lung.

SISTEMAS DE ENSAMBLAJE PARA EL ESTUDIO Y CARACTERIZACIÓN DE LA RELACIÓN ESTRUCTURA-FUNCIÓN DE PROTEÍNAS DEL SURFACTANT PULMONARY SP-B Y SP-C
RESUMEN
El surfactante pulmonar es un complejo lipoproteico encargado de disminuir la tensión superficial en la interfase aire-líquido alveolar. La proteína del surfactante pulmonar C (SP-C) es una proteína transmembrana de 35 aminoácidos, muy conservada. Participa en el transporte de moléculas tensioactivas para reducir la tensión superficial en la interfase aire-líquido pulmonar. Dada su alta hidrofobicidad, en este trabajo se presentan dos sistemas de nanocomplejos que permiten su solubilización. Éstos consisten en parches de membrana rodeados, bien por dos copias de la proteína de andamiaje MSP1D1, o por un copolímero de estireno-ácido maléico SMA. Se han realizado estudios biofísicos con ambos sistemas en presencia o no de SP-C nativa o de una versión recombinante humana (rSP-C). Los nanodiscos de MSP1D1 tienen actividad interfacial similar en ausencia y presencia de proteína, e inhiben la actividad del surfactante. Los nanodiscos de SMA en presencia de ambas proteínas parecen acelerar la adsorción de los lípidos a la interfase, alterando mínimamente la función del surfactante. Aunque estos nanodiscos parecen anclarse a las membranas fosfolipídicas más fuertemente que los nanodiscos vacíos, ambos son capaces de excluirse de la interfase, lo que resulta interesante para su posible uso como sistemas de transporte de fármacos al interior pulmonar.

PROTEINS FROM A BIOCHEMICAL POINT OF VIEW
ABSTRACT
The structural and functional study of proteins at the molecular level requires high yield and quality
systems of production and purification. The researcher Lucía García Ortega is focused on the
development of heterologous systems to obtain recombinant proteins which match these properties
with the ultimate purpose of understanding the molecular determinants involved in their function.
Part of her work is centered in the study of fungal toxic proteins with ribonucleolytic activity against
the ribosome, the ribotoxins. Different strategies have been achieved from the study of the recognition
mechanism of the ribosome by the ribotoxins to their insecticide ability and their potential
biotechnological applications. In addition, a new line of research is focused on the study of saposins
which are lipid-interacting proteins. In particular, those associated to the pulmonary surfactant that is
a protein-lipid mixture at the surface of alveoli, involved in respiratory mechanics and innate immune
defense against numerous pathogens.

RESUMEN
El estudio funcional y estructural de proteínas a nivel molecular requiere de sistemas de producción y
purificación de éstas con alto rendimiento y calidad. La investigadora Lucía García Ortega se ha
especializado en el uso de sistemas heterólogos para la obtención de proteínas recombinantes que
cumplan estos requisitos, con el fin de poder analizar en detalle los determinantes moleculares
implicados en su función. Parte de su trabajo se centra en el estudio de proteínas tóxicas procedentes
de hongos con actividad ribonucleolítica, las ribotoxinas, cuyo sustrato natural es el ribosoma. En este
campo, se realizan estudios con distintos enfoques, desde el modo de reconocimiento del ribosoma
por parte de las ribotoxinas a nivel molecular, hasta la capacidad insecticida de éstas y sus potenciales
aplicaciones biotecnológicas. Por otro lado, se está comenzando una nueva línea de trabajo enfocada
en el estudio de proteínas de tipo saposina, con capacidad de interaccionar con lípidos. En particular,
aquellas que se encuentran en el entorno del surfactante pulmonar, que es una mezcla lipoproteica
que, en la superficie de los alveolos, permite la función respiratoria y está implicada en la defensa
innata frente a patógenos respiratorios.

TISSUE-SPECIFIC FACTORS NEEDED FOR TISSUE REPAIR AND TYPE 2 ACTIVATION OF
MACROPHAGES
ABSTRACT
Stimulation of macrophages with interleuquin-4 (IL-4) is essential for tissue repair, since it induces the
production of mediators to promote cell proliferation and extracellular matrix production. However,
exuberant alternative (type 2) activation of macrophages is associated to the development of diseases
such as fibrosis, asthma and even cancer. We have recently discovered that tissue-specific factors
produced in the local environment are required for complete alternative activation of tissue
macrophages induced by IL-4 (Minutti et al., Science 2017). In the lung, the amplifying factor is
surfactant protein A (SP-A), whereas in the liver and peritoneal cavity, the enhancing molecule is C1q,
a protein structurally similar to SP-A. IL-4 increases the production of SP-A by alveolar epithelial cells,
and C1q by myeloid cells, as well as membrane expression of their receptor Myo18A, which mediates
enhancing effects of SP-A or C1q on IL-4-stimulated macrophages, leading to an amplification loop of
IL-4 signalling. We have shown that amplification of IL-4 signalling in macrophages is beneficial for
efficient control of infection and tissue repair. SP-A promotes lung tissue repair in a murine model of
infection with the parasite Nippostrongylus brasiliensis, and C1q favors liver tissue regeneration in a
model of Listeria monocytogenes infection. However, by amplifying alternative activation of
macrophages, C1q worsened fibrosis symptoms of peritoneal fibrosis in mice. Deciphering the factors
and conditions that modulate alternative activation of macrophages will help to discover new
treatments for pathologies with a type 2-biased immune response, such as fibrosis.

RESUMEN
La estimulación de macrófagos con interleuquina 4 (IL-4) tiene un papel esencial en la reparación
tisular, ya que induce la secreción de mediadores para la proliferación celular y la producción de matriz
extracelular. Por otro lado, una activación de macrófagos por IL-4 (tipo 2 o alternativa) descontrolada
está relacionada con patologías como la fibrosis, el asma o incluso el cáncer. Recientemente, hemos
demostrado que existen factores locales, específicos de tejido, necesarios para la completa activación
de macrófagos por IL-4 (Minutti et al., Science 2017). En el pulmón, la molécula amplificadora de la
señal de IL-4 es la proteína del surfactante A (SP-A), mientras que en el hígado y en la cavidad
peritoneal, el amplificador local es la proteína C1q, estructuralmente similar a SP-A. IL-4 incrementa la
producción de SP-A y C1q en dichos tejidos, así como la expresión en la superficie de macrófagos del
receptor Myo18A, que media el efecto amplificador de SP-A y C1q sobre la activación por IL-4, dando
lugar a un bucle de amplificación de la señal de IL-4 en cada tejido. Hemos demostrado que este
sistema de amplificación de la señal IL-4 es beneficioso para combatir la infección y reparar el órgano
dañado. Así, la SP-A promueve la reparación del pulmón tras la infección con el parásito
Nippostrongylus brasiliensis y, en el hígado, C1q favorece la regeneración tisular en un modelo de
infección con la bacteria Listeria monocytogenes. La otra cara de la moneda es que C1q, al potenciar
la activación tipo 2 de macrófagos, empeoró la fibrosis peritoneal en un modelo de diálisis peritoneal
en ratones. Actualmente, continuamos descifrando las claves que modulan la activación alternativa de
macrófagos para descubrir qué condiciones contribuyen al desarrollo de patologías como la fibrosis,
lo que permitirá el diseño de nuevos tratamientos.

OPTIMIZATION OF A PRECLINICAL THERAPY OF CANNABINOIDS IN COMBINATION
WITH TEMOZOLOMIDE AGAINST GLIOMA
ABSTRACT
Glioblastoma multiforme (GBM) is the most frequent and aggressive form of brain cancer. These
features could be explained by the presence within the tumor mass of a small population of cells
termed Glioma Initiating Cells (GICs) that has been proposed to be responsible for the relapses
occurring in this disease. Thus, the development of novel therapeutic approaches (specifically focused
in the elimination of GICs) is urgently needed to improve the survival of patients with this devastating
disease.
Δ9-Tetrahydrocannabinol (THC, the major active ingredient of marijuana), and other cannabinoids (for
example cannabidiol, CBD) have been shown to exert antitumoral actions in several animal models of
cancer, including glioma. The mechanism of these anticancer actions relies, at least in part, on the
ability of these compounds to stimulate autophagy-mediated apoptosis in tumor cells. With the aim
of optimizing the possible clinical utilization of cannabinoids in anti-GBM therapies, we explored the
anticancer efficacy of the systemic administration of cannabinoids in combination with temozolomide
(TMZ is the benchmark agent for glioma treatment) in preclinical models of glioma. Our results show
that oral administration of Sativex-like extracts (containing THC and CBD at a 1:1ratio) in combination
with TMZ produces a strong antitumoral effect in both subcutaneous and intracranial glioma cell-
derived tumor xenografts even in those tumors derived of GICs obtained from biopsies of patients. In
those tumors generated by GICs injection, a high proportion of CBD in the cannabinoid extracts showed
therapeutical benefits compared to THC: CBD at 1:1 ratio.
These findings support that the administration of TMZ along with combinations of THC: CBD, and
specifically those containing a higher proportion of CBD could be exploited therapeutically for the
treatment of patients with GBM, affecting not only to the vast majority of more differentiated tumor
cells but also to the population of GICs.

OPTIMIZACIÓN DE UNA TERAPIA PRECLÍNICA DE CANNABINOIDES EN COMBINACIÓN
CON TEMOZOLOMIDA EN GLIOMAS
RESUMEN
El glioblastoma multiforme (GBM) es la forma más frecuente y agresiva de cáncer cerebral. Esta
extremada agresividad se explica por la presencia dentro de la masa tumoral de una pequeña
población de células denominadas células iniciadoras de glioma (GICs). Se propone que estas células
son las responsables de las recaídas que se producen en la práctica totalidad de estos enfermos. Así,
se necesitan con urgencia nuevas estrategias terapeúticas (y específicamente aquellas dirigidas a la
población de GICs) para mejorar la supervivencia y calidad de vida de los pacientes.
El Δ9-tetrahidrocannabinol (THC, el principal ingrediente activo de la marihuana), y otros
cannabinoides tales como el cannabidiol (CBD) han demostrado tener propiedades antitumorales en
modelos animales de cáncer, incluyendo los gliomas. El mecanismo de estos efectos en cáncer es
debido, al menos en parte, a la habilidad que tienen estos compuestos para inducir un proceso de
autofagia en las células tumorales que posteriormente desencadena la muerte por apoptosis de dichas
células. Con el objetivo de optimizar una posible utilización clínica de los cannabinoides en terapias
contra el glioblastoma, exploramos la eficacia de la administración sistémica de cannabinoides en
combinación con Temozolomida (TMZ, el agente quimioterapeútico de referencia para el tratamiento
de los gliomas) en modelos preclínicos de esta enfermedad. Nuestros resultados muestran que la
administración oral de extractos de cannabinoides con composición similar al fármaco Sativex
(contiene THC y CBD en proporciones 1:1) en combinación con TMZ ejerce una acción antitumoral
mucho más pronunciada que los tratamientos simples tanto en tumores subcutáneos como
intracraneales, incluso en aquellos tumores generados a partir de GICs obtenidas de biopsias de
pacientes. Además, en el caso de estos tumores generados con GICs se ha observado que el
incremento de la proporción de CBD en el fármaco cannabinoide potencia el efecto antitumoral.
Estos hallazgos apoyan que la administración de TMZ junto con combinaciones de THC: CBD, y
específicamente aquellas que contienen una mayor proporción de CBD podrían explotarse
terapéuticamente para el tratamiento de pacientes con GBM incidiendo de manera eficaz no sólo en
la población de células del tumor más diferenciada sino también en la población de GICs.

IMMUNOLOGICAL MECHANISMS INVOLVED IN THE MODE OF ACTION OF NEW
VACCINES DIRECTED AGAINST DENDRITIC CELLS
ABSTRACT
We are interested in the study of the functional role played by human dendritic cells (DCs) in
inflammatory processes, immune tolerance related diseases, infectious diseases as well as in the
restauration of healthy immune responses after immunotherapy. Our work is focused on the study of
the immunological mechanisms underlying the clinical benefit of different types of vaccines employed
for allergen-specific immunotherapy (AIT), for the treatment and prevention of recurrent infections
affecting the respiratory or urinary tracts and other immune tolerance related diseases, such as cancer.
One of the main novel lines of research of the group is focused on studying the role played by
cannabinoids and other small molecules in the immunomodulation of human DCs in the context of
allergic diseases. We are also interested in the elucidation of the molecular mechanisms involved in
food allergic sensitization. Overall, we aimed at the identification of molecules in human DCs as
potential therapeutic targets to develop novel vaccines and alternative immunotherapy protocols for
the treatment of immune tolerance related diseases and infections.
Main lines of research:
1. Study of the role played by the endocannabinoid system in the context of allergic diseases focusing
on the immunomodulation exerted by cannanbinoids in human DCs.
2. Vaccine development and study of the immunological mechanisms of action in:
i). Allergy. Allergoids conjugated to mannan as next generation vaccines able to generate healthy
immune responses to allergens (see graphical abstract)
ii). Infectious diseases (polybacterial vaccines to prevent and treat recurrent infections).
3. Study of the immunomodulatory properties of novel carbohydrates involved in cancer in human
dendritic cells.
4. Study of the immunological mechanisms governing food allergic sensitization and the actual role
played by human DCs, Th2 and Treg cells in such processes.

VACUNAS DIRIGIDAS FRENTE A CÉLULAS DENDRÍTICAS
RESUMEN
Nuestro grupo de investigación está interesado en el estudio del papel funcional que desempeñan las
células dendríticas (DCs) humanas en procesos inflamatorios y en la restauración de respuestas no
patológicas tras tratamientos de inmunoterapia. Además, trabajamos en la identificación de nuevas
dianas terapéuticas, en la identificación y caracterización de nuevos alérgenos de alimentos y en el
estudio de los mecanismos inmunológicos implicados en alergia.
Nuestro trabajo se centra en el estudio de los mecanismos inmunológicos asociados al modo de acción
de nuevas vacunas dirigidas frente a DCs para el tratamiento de la alergia (alergoides conjugados a
manano), para la prevención y tratamiento de enfermedades recurrentes del tracto respiratorio y
urinario (vacunas polivalentes bacterianas enteras inactivadas por calor). Asimismo, estamos
interesados en el estudio del papel inmunodulador sobre DCs humanas de carbohidratos implicados

inmunomodulador de los cannabinoides en DCs humanas en el contexto de enfermedades alérgicas

ROLE OF ENDOCANNABINOID SYSTEM IN BREAST CANCER
ABSTRACT
Breast cancer is one of the leading causes of death amongst women. Although the mortality rates
associated with this disease are decreasing, important clinical challenges remain unsolved, such as
innate or acquired resistance to conventional treatments, a better stratification of the patients to
determine which are the optimal therapies in each particular case, and reducing the side effects of
their current treatments, which in many cases have a significant negative impact in their quality of life.
It has been demonstrated that cannabinoids have therapeutic properties of interest for cancer patients
(they prevent nausea and vomiting induced by chemotherapy, they are analgesic, anxiolytic and
appetite stimulant, etc.). Our research, however, focuses on the antitumor potential of these
compounds, which have been shown to induce antiproliferative, antiangiogenic and antimetastasic
effects in different preclinical models of cancer.
In this context, our main aims are:
(i) Understanding the role of the endocannabinoid system in breast cancer generation and progression.
Our work has demonstrated, for example, that cannabinoid receptor CB2R plays a pivotal role in HER2
pro-oncogenic signaling, and that CB2R expression may be used as a prognostic marker in HER2+ breast
cancer.
(ii) Analyzing the role of the endocannabinoid system as a target for new antitumor treatments for the
management of HER2+ breast cancer. Our work has demonstrated, for example, that cannabinoids
produce antitumor responses in animal models of the different subtypes of breast cancer.
Our final aim is to translate this preclinical knowledge into clinical tools for the treatment of breast
cancer.

RESUMEN
El cáncer de mama es una de las principales cause de muerte entre mujeres. Aunque la mortalidad
asociada a esta patología está en descenso, existen todavía desafíos clínicos a los que tenemos que
dar respuesta, tales como la resistencia a los tratamientos actuales (tanto innata como adquirida), una
mejor subclasificación de las pacientes, para determinar cuál es el mejor tratamiento en cada caso, y
la reducción de los efectos secundarios que producen los tratamientos actuales, que en muchos casos
disminuyen notablemente la calidad de vida de las pacientes.
Se ha demostrado que los cannabinoides tienen propiedades terapéuticas muy interesantes para los
pacientes oncológicos (previenen las náuseas y los vómitos inducidos por quimioterapias, son
analgésicos, ansiolíticos y estimuladores del apetito, etc.). Nuestra investigación, sin embargo, se
centra en el estudio del potencial antitumoral de estos compuestos, que producen respuestas
antiproliferativas, antiangiogénicas y anti-metastásicas en distintos modelos preclínicos de cáncer.
En este contexto, nuestros principales objetivos son:
(i) Entender el papel que desempeña el sistema endocannabinoide en la generación y progresión del
cáncer de mama. Nuestro trabajo ha demostrado, por ejemplo, que el receptor de cannabinoides CB2R
es una pieza fundamental en la maquinaria de señalización pro-oncogénica de HER2, y que su
expresión podría utilizarse como marcador pronóstico en cáncer de mama HER2+.
(ii) Analizar el potencial del sistema endocannabinoide como diana de tratamientos antitumorales en
cáncer de mama. Nuestro trabajo ha demostrado, por ejemplo, que los cannabinoides producen
respuestas antitumorales en modelos animales de los distintos subtipos de cáncer de mama.
El objetivo final de nuestra investigación es trasladar todo este conocimiento preclínico al entorno
clínico para analizar si las pacientes con cáncer de mama pueden incorporar los cannabinoides a sus
tratamientos antitumorales.

COMPUTATIONAL MODELS OF REGULATORY AND METABOLIC EVOLUTION OF
COMPLEXITY
ABSTRACT
Under unlimited availability of resources, cells are able to evolve a range of functions as a result of
adaptation to their environment, without necessarily reaching extreme efficiencies in the
management of material sources. Conversely, when such sources are limited, satisfying multiple
simultaneous environmental constraints is possible only under strict regulation. At the same time,
living beings face fast environmental changes during their lifetime, e.g. throughout yearly seasons,
whose time scales are generally much shorter than those of evolution. In such changing environments,
regulation becomes essential. Our work explores the effect of both environmental and material
constraints on the evolution of regulation. This is achieved by means of simple models with the
following features. First, environmental constraints are represented by means of concentration
thresholds of certain cell products, which must be reached in order for a function to be effective.
Secondly, all cell products are synthesized from a single material source, whose flux is considered
constant and limited. Finally, the synthetic flux of each product may be influenced, by virtue of
genetically-codified kinetics, by the presence of other products and by environmental constraints. Our
results suggest that the environmental pressure leads to low levels of superfluous products, even
though selection does not act directly on them. Additionally, Darwinian evolution is, per se, inefficient,
and often unable, to generate the required regulation, unless sufficiently wide neutral phenotype
spaces exist. The latter result seems to be a consequence of epistasis between the various genetic
products.

RESUMEN
La disponibilidad ilimitada de recursos permite a las células la evolución simultánea de múltiples
funciones como resultado de la adaptación al medio, sin necesidad de alcanzar una gran eficiencia en
el uso de materias primas. Sin embargo, cuando estas son limitadas la satisfacción de varios requisitos
ambientales simultáneos solo es posible bajo una estricta regulación. Al mismo tiempo, los seres vivos
se enfrentan a cambios ambientales rápidos en el transcurso de su vida (por ejemplo, en distintas
estaciones del año), en escalas temporales mucho más cortas que las propias de la evolución. En tales
entornos cambiantes, la regulación resulta también esencial. En este trabajo se exploró el efecto tanto
de las restricciones ambientales como de los materiales sobre la evolución de la regulación, mediante
un modelo sencillo con las siguientes características. Primero, las restricciones ambientales se
representan como umbrales de concentración sobre determinados productos celulares, umbrales que
se deben sobrepasar para que la adaptación se considere efectiva. En segundo lugar, todos los
productos celulares se sintetizan a partir de una única fuente de material, cuyo flujo se considera
constante y restringido. Por último, el flujo de síntesis de cada producto se puede ver afectado,
mediante una cinética codificada genéticamente, por la presencia de otros productos y de las
restricciones ambientales. Los resultados obtenidos mediante dicho modelo indican que la presión
ambiental conduce a niveles bajos de productos superfluos, a pesar de que sobre ellos no actúa una
presión selectiva que los restrinja directamente. Por otra parte, la evolución darwiniana estricta es
ineficiente, a menudo incapaz, de generar por si sola la regulación necesaria, que solo aparece cuando
existen espacios fenotípicos neutrales suficientemente amplios. Esto último parece ser debido a la
epistasis entre los distintos productos génicos.

THERMODYNAMIC STUDY FOR THE SIMULATION OF SOLVENT-POLYMER
PURIFICATION SYSTEMS
ABSTRACT
Nowadays polymeric materials are extremely important due to their wide range of applications. The
final use of these materials will depend on how they are produced and how they are purified after the
polymerization. As these processes are usually carried out by steam stripping or devolatilization, it is
extremely important to deeply study the thermodynamic of the mixtures involving the polymer, the
organic solvent, the monomers and water. Therefore, the aim of this research is to perform a rigorous
thermodynamic assessment to the mixtures involved in the purification of SBS and EVA copolymers.
The first thermodynamic model we employed was the well-known Flory-Huggins equation, calculating,
for each polymer-solvent mixture, its binary interaction parameter by means of Intrinsic Viscosity and
Inverse Gas Chromatography techniques. However, we noticed that the obtained values with both
techniques were quite different so it is important to consider the dependence of the Flory Huggins
parameter of composition. So, based on the previous works of Wolf [1], we have developed and
validated a methodology which allows, from only two values of the Flory Huggins parameter of a
certain polymer-solvent mixture, obtain its values in the entire range of solvent composition (from 0
to 1). With these values, it is possible to obtain the pressure-composition equilibrium curve at constant
temperature, which can be efficiently fitted to whatever thermodynamic model, such as PC-SAFT [2].

RESUMEN
Hoy en día los materiales poliméricos son de gran importancia debido a su amplio rango de
aplicaciones. Sin embargo, el uso final de muchos de ellos va a depender de cómo se obtienen y de
cómo se purifican después de la polimerización. Esta última etapa se suele llevar a cabo mediante
arrastre con vapor de agua o evaporación directa por lo que es de gran importancia llevar a cabo un
estudio termodinámico riguroso de las mezclas que involucran al polímero, al disolvente orgánico, a
los monómeros y el agua. Así, el objetivo de esta investigación es llevar a cabo un estudio
termodinámico riguroso de las mezclas que aparecen en los procesos de purificación de los
copolímeros SBS y EVA. El primer modelo termodinámico que utilizamos fue la conocida ecuación de
Flory-Huggins; para ello, para cada mezcla binaria polímero-disolvente, calculamos el parámetro de
interacción binaria mediante Viscosimetría Intrínseca y mediante Cromatografía Inversa de Gases. Sin
embargo, los valores obtenidos por ambas técnicas eran bastante diferentes por lo que concluimos
que es importante considerar la dependencia de este parámetro de interacción binaria de Flory-
Huggins con la composición. Basándonos en los trabajos previos de Wolf [1] hemos desarrollado y
validado una metodología que permite, partiendo únicamente de dos valores del parámetro de
interacción binaria para una determinada mezcla polímero-disolvente, obtener los valores de dicho
parámetro en todo el intervalo de composiciones. Con estos valores, se puede obtener de manera
sencilla la curva de equilibrio presión-composición a temperatura constante, cuyos datos se pueden
ajustar de manera rigurosa a cualquier modelo termodinámico, como PC-SAFT [2].

FORMULATION AND CHARACTERIZATION OF IONOGELS
ABSTRACT
The research line presented at the 1st Young Researchers Conference celebrate at the Faculty of
Chemical Sciences is part of the lines investigated in the UCM Group "Development of processes and
chemical products with low environmental impact"; therefore, this line is linked to some of the other
research of the group, such as biorefinery and the employ of ionic liquids as solvents and in the
development of new environmentally-friendly processes. This line of research aims to the
development of new materials, specifically in gels formulated using ionic liquids (ILs) and cellulose as
matrix and reinforced using chitosan with different degrees of deacetylation; these gels are therefore
named ionogels. Among the employed ionic liquids, the so-called third generation of ILs are currently
used in the line, specifically choline-based ILs. Thus, given the properties of these ionogels, wed
decided to denote them as bionogels. Some of their main applications are focused on the
medical/pharmacological sector. Once formulated, these ionogels are characterized, with special
emphasis on the determination of their thermal and rheological properties, as well as in their
biodegradability and toxicity.

La línea de investigación expuesta en las primeras Jornadas de Jóvenes Investigadores de la Facultad
de Ciencias Químicas forma parte de las líneas de investigación del Grupo UCM consolidado
“Desarrollo de procesos y productos químicos de bajo impacto ambiental”, de forma que enlace con
otras líneas de investigación del grupo como son la biorrefinería y el empleo de líquidos iónicos como
disolventes y en el desarrollo de procesos. Esta línea de investigación aborda el desarrollo de unos
nuevos materiales compuestos, en concreto en el desarrollo de geles formulados empleando líquidos
iónicos (LIs) con celulosa como matriz y reforzados y quitosanos con distintos grados de desacetilación,
por lo que estos geles se denominan comúnmente ionogeles. Dentro de los líquidos iónicos utilizados,
los llamados de tercera generación son lo que actualmente se están empleando en la línea, en concreto
LIs derivados de la colina. De este modo, dadas las propiedades de estos ionogeles, se ha optado por
denominarlos de forma particular como bionogeles. Algunas de sus principales aplicaciones se centran
en el sector médico/farmacológico. Una vez formulados, estos ionogeles son caracterizados, poniendo
especial énfasis en la determinación de sus propiedades térmicas y reológicas, así como en su
biodegradabilidad y toxicidad.

ENVIRONMENTAL CHEMICAL ENGINEERING: SUSTAINABLE PRODUCTION OF
RECYCLED PAPER AND ADVANCED WASTEWATER TREATMENT
ABSTRACT
The objective of this presentation is to review some of the studies carried out in the last decade
applying environmental chemical engineering into sustainable production of recycled paper and
advanced wastewater treatment.
First, sustainable recycled paper production was analyzed in terms of water and waste management.
Many projects have been carried out to minimize the fresh water consumption in the paper industry
by reuse of process water by internal treatments, reuse of the effluent and the use of reclaimed water.
All these studies have been carried out in close cooperation with the paper industry (Holmen Paper
Madrid), this paper mill becoming the first paper industry in Europe using 100% reclaimed water. In
the area of waste management, several studies have been carried to demonstrate the technical
feasibility of recycling deinking sludge in different applications such as agricultural use and
incorporation in building materials, e.g. fibercement, mortars and cement.
Second, the presentation describes some on-going research within the research group. One project
named “CONCENTRADOS-SOS” deals with the treatment of concentrates (or rejects) from different
industrial sectors aiming to extend the limits of water reuse and sustainable water use. First results,
treating raw landfill leachates and concentrates from landfill leachates were presented. Other
important area in the research group is the obtention and application of nanocelluloses in different
industrial sectors, including paper industry, fiber-cement industry and different environmental
applications (flocculants, adsorbents, membranes, etc.).

residuos producidos. Se han llevado a cabo muchos proyectos para minimizar el consumo de agua de
alimentación en la industria papelera mediante la reutilización del agua de proceso con tratamientos
internos, la reutilización del efluente o el uso de agua regenerada. Todos estos estudios se han llevado
a cabo cooperando estrechamente con la industria papelera (Holmen Paper Madrid), convirtiéndose
esta compañía en la primera fábrica de papel en Europa usando 100% agua regenerada. En el área de
la gestión de residuos, se han llevado a cabo una serie de estudios para demostrar la viabilidad técnica
del reciclaje de los lodos de destintado en diferentes aplicaciones como el uso en agricultura o su
incorporación en materiales de construcción, p.ej. fibrocemento, morteros y cemento.
En segundo lugar, la presentación describe algunos de los proyectos que se están llevando a cabo
actualmente en el grupo de investigación. Un Proyecto denominado “CONCENTRADOS-SOS” versa
sobre el tratamiento de concentrados (o rechazos) de diferentes sectores industriales con el objetivo
de extender los límites de la reutilización de agua y el uso sostenible del agua. En la presentación se
recogen los primeros resultados obtenidos en el tratamiento de lixiviados y concentrados de lixiviados
de vertedero. Otra área importante del grupo de investigación es la obtención y aplicación de
nanocelulosas en diferentes sectores industriales, que incluyen la industria papelera, la industria del
fibrocemento y diferentes aplicaciones medioambientales (floculantes, adsorbentes, membranas,
etc.)

PLASMA ELECTROLYTIC OXIDATION OF LIGHT ALLOYS
ABSTRACT
The demand of modern technological society for light structural materials (Al, Ti, Mg) emphasizes a
combination of good corrosion resistance with wear properties and functionalized surfaces. Their
extensive field of applications ranges from mechanical aspects and transport components to
bioengineering. Regardless of the final application, improved tailored surfaces are required to prolong
service life and reduce long-term costs. Plasma Electrolytic Oxidation (PEO) is an exceptional candidate
to achieve that goal: it enables a considerable improvement of the mechanical properties and
corrosion resistance of light alloys together with other characteristics such as improved
biocompatibility. PEO is an environmentally friendly treatment process employed to produce relatively
thick (10-100µm) ceramic-like coatings on Mg, Al, Ti and other valve metals, with incorporation of
species from both the substrate and the electrolyte. The coatings are formed under high voltages,
exceeding those of dielectric breakdown, when short-lived discharges occur locally on the coating
surface and the current density and temperature are greatly increased, facilitating the formation of
phases normally associated with processing at relatively high temperatures. The current research lines
compile studies of PEO-coated light alloys for structural (Al, Mg) and biomedical applications (Ti, Mg)
for corrosion protection and surface functionalization.

OXIDACIÓN ELECTROLÍTICA POR PLASMA DE ALEACIONES LIGERAS
RESUMEN
El campo de aplicación de las aleaciones ligeras (Magnesio, Aluminio y Titanio) es muy extenso y abarca
desde los medios de transporte a la biomedicina.
Sin embargo, estos materiales necesitan una mejora en aspectos de corrosión, desgaste y/o
biocompatibilidad dependiendo de su aplicación final. La modificación superficial mediante
recubrimientos por oxidación electrolitica por plasma (OEP) es una solución a estas necesidades ya
que generan una capa de óxido que puede adaptarse en forma y composición dependiendo de la
aleación y su campo de aplicación:
1. En la industria del transporte y con el objetivo de disminuir el peso de los vehículos, los
recubrimientos OEP se aplican sobre Mg y Al para mejorar su comportamiento a corrosión y desgaste.
Además, estos recubrimientos permiten la incorporación de inhibidores, dándoles capacidad auto-
curativa una vez que se ha producido un defecto.
2. En el campo de los biomateriales se pueden generar recubrimientos OEP ya sea para implantes
permanentes (Titanio) o biodegradables (Magnesio) con una biocompatibilidad de superficie mejorada
debido a la incorporación de Ca y P, obteniendo una composición final cercana a la hidroxiapatita,
compuesto mayoritario en los huesos.

INTEGRATED BIOREFINERY. ENERGY CROPS VALORIZATION
ABSTRACT
The ultimate objective of the research programme is to know the energy conversion potential of
Cynara cardunculus L. for cultivation in the Region of Madrid, Spain. This programme covers a wide
range of research areas including prospective GIS assessment, agronomy, biomass pre-treatments,
fluidized bed combustion, fluidized bed air-gasification, heterogeneous catalysis for biodiesel
production, catalytic materials for valorization of glycerol via propanodiols, valorization of seed oil cake
via furfural, valorization of side-products and by-products of the energy chain to functional ingredients.
UCM research group carried out some gasification tests, at pilot scale, in order to evaluate the
technical feasibility of the process, using a facility consisting of a bubbling fluidized bed gasifier and a
catalytic gas cleaning system. The catalytic cleaning unit selected in this process is a second-generation
monolithic reactor, containing two pieces of monoliths used in its commercial size (15 x 15 x 30 cm).
The main results obtained showed promising perspectives in the valorization of cardoon by means of
air gasification process. The L.H.V. values of the fuel gas were around 5.8-6.5 MJ/Nm3db. Tar
conversion as high as 88 % was achieved in the catalytic reactor.

BIORREFINERIA INTEGRADA. VALORIZACION DE CULTIVOS AGROENERGETICOS
RESUMEN
El principal objetivo de la línea de investigación es conocer la conversión energética potencial del
Cynara cardunculus L. para su cultivo en la región de Madrid. Esta investigación se englobaría dentro
del concepto de biorrefinería, cubriendo un gran rango de áreas de investigación como la valorización
del suelo mediante GIS (“geographical information system”), agronomía, pretratamientos de la
biomasa, combustión y gasificación en lecho fluidizado, catálisis heterogénea para la producción de
biodiesel, materiales catalíticos para la valorización de glicerol vía propanodioles, valorización de la
torta de extracción del aceite de la semillas vía furfural, valorización de subproductos de la cadena
energética para la producción de ingredientes funcionales.
El grupo de investigación de la UCM llevó a cabo ensayos de gasificación con aire del cardo, a escala
de planta piloto, para evaluar la viabilidad técnica del proceso, utilizando una instalación con un
reactor de lecho fluidizado burbujeante seguido de un reactor catalítico de limpieza del gas de
gasificación. La unidad de limpieza catalítica, es un reactor de monolitos de segunda generación que
alberga dos piezas de catalizador en su tamaño comercial (15 x 15 x 30 cm).
Los principales resultados obtenidos son optimistas en la valorización energética del cardo por medio
de procesos de gasificación. EL PCI (poder calorífico inferior) del gas de gasificación obtenido presenta
unos valores entre MJ/Nm3db. La conversión en la eliminación de alquitranes en el reactor catalítico
alcanzó valores hasta del 88 %

Muchos de los científicos de este país se forman en la Universidad y muchos de
ellos continúan su carrera científica como parte de las plantillas de Centros de
investigación, nacionales e internacionales, donde se aprovecha su excelente
formación científica. Sin embargo, es obligación de la UCM tratar de recuperar a
estos jóvenes científicos para que puedan revitalizar a nuestra Institución
aportando sus conocimientos, su juventud, y su empuje. De hecho, la atracción de
todo este talento ha permitido a nuestra Facultad desarrollar una investigación
puntera y de gran calidad. Sin embargo, es verdad que a veces hacemos más
publicidad de los de fuera de nuestra Comunidad Universitaria que de nuestros
propios científicos.
Desde la Facultad de Ciencias Químicas de la Universidad Complutense se ha
realizado un proyecto en forma de un ciclo de conferencias durante el curso
académico 2018/2019 con el título: ¿Qué investigan los jóvenes científicos de la
Facultad de Ciencias Químicas?, cuyo objetivo es hacerles visibles en la Comunidad
Científica, divulgar y trasmitir los resultados de sus investigaciones y servir de
referentes para estimular las vocaciones científicas entre nuestros alumnos.
