HISTORIA CRONOLÓGICA DEL DNA
• 1865 Gregor Mendel – presenta sus trabajos donde explica
como se transmiten los rasgos y que patrones muestran.
Presenta sus 3 principios o Leyes:
> Ley de Dominancia
> Ley de Segregación
> Ley de Sorteo Independiente
• 1869 Friedrich Miescher purifica el DNA de células y llama a
este material “nucleína”.
• 1888 Se identifica el material del núcleo y se le llama
cromosomas.
• 1903 Walter Sutton identifica a los cromosomas como los
agentes físicos que llevan los “factores” (genes) que
mencionaba Mendel.

1928 Fred Griffith – realiza experimentos con cepas de
bacterias que causan pulmonía (Ver Fig. 13.2 ) e identifica
un “factor transformador” que podía alterar los rasgos de
las cepas bacterianas.
1932 Robert Feulgen - identifica al DNA como el material
presente en los cromosomas.
1940 Erwin Chargaff – estudia la composición de los
nucleótidos del DNA y encuentra que los % de nucleótidos
de timina son iguales a los de adenina y que los % de
nucleótidos de citosina son iguales a los de guanina.
1944 Oswald Avery, Colin McLeod y Maclyn McCarty
identifican al “factor transformador de Griffith.como el DNA.

1950’s Alfred Hershey y Martha Chase realizan experimentos con bacteriofagos marcando con radioisótopos de fósforo al DNA y de azufre a las proteínas . Demostraron que era el DNA de los bacteriofagos el que entraba a las bacterias y no las proteínas corroborando que el “factor transformador” era el DNA (Ver diagrama de los experimentos).
1951 Rosalind Franklin – estudia el arreglo de átomos en la molécula de DNA utilizando cristalografía de rayos X, pero su interpretación no fue correcta.
1953 James Watson y Francis Crick – proponen un modelo químico y físico de la molécula de DNA utilizando la información de Franklin. Se les otorga el Premio Nobel en química por su trabajo.

FUNCIONES BÁSICAS DEL DNA
Almacenaje de la Información Genética
La información genética se encierra en la secuencia de los 4 nucleótidos,
la cual dicta la secuencia de aminoácidos en un polopéptido.
Autoreplicación y Herencia
La replicación del DNA es semiconservativa y ocurre previo a que se
divida la célula. De esta forma la información genética se transfiere de
una célula “madre” a una célula “hija” y así sucesivamente.
Expresión del Mensaje Genético
La información encerrada en el DNA se transfiere a una molécula de RNA
en lo que se conoce como transcripción. Luego este mensaje se traduce
en secuencia de aminoácidos en un polipéptido. Finalmente este
polipéptido se expresa en forma de un rasgo particular.

SEMICONSERVATIVE DNA REPLICATION IN
BACTERIA
Each strand of a DNA double helix is a template for synthesis of a complementary strand of DNA.
One template builds DNA continuously; the other builds DNA discontinuously, in segments.
Each new DNA molecule consist of one old strand and one new strand.
The bubble that initiate replication of the circular bacterial chromosome forms at a genetically specified point in the chromosome called the origin.
Then the 2 replication forks created by the bubble travel simultaneously in opposite directions around the circular chromosome .
Eventually they meet halfway around the chromosome at a point called the terminus.

ENZYMES OF DNA REPLICATION IN BACTERIA
REPLICATION OF THE LEADING STRAND
DNA helicase - Breaks hydrogen bonds between DNA strands separating the strands of the old DNA and moves the replication fork.
Single-strand binding protein(SSBP) - Binds to the exposed single strands of DNA and keeps them separate during replication.
DNA polymerase III - Joins free nucleotides into a new strand of DNA.
REPLICATION OF THE LAGGING OR DISCONTINUOUS STRAND
Primase - Makes a a short RNA primer, then the DNA polymerase III adds DNA to the RNA primer as it moves away from the replication fork and continues until it runs into the 5’end of a previous RNA primer.
DNA polymerase I- It destroys the RNA primer while replacing it with DNA .
DNA ligase - Joins DNA segments on discontinuous strand sealing the gap between the newly synthesized fragment of DNA and the continuous strand in front of it.

REGULACION DE LA EXPRESIÓN GENÉTICA
La expresión genética de una bacteria es usualmente regulada aumentando o disminuyendo la razón de transcripción o traducción.
Las enzimas inducidas se producen solamente cuando su sustrato es abundante. Las enzimas reprimidas (“repressible”) se producen solamente cuando su producto es escaso. Las enzimas constitutivas se producen siempre.
La transcripción se regula usualmente por proteínas que se pegan al DNA y cambian su interacción con la polimerasa de RNA. Si se producen pocos transcritos se produce poca proteína.
Un operón es un set de genes que se regulan y transcriben juntos. El operón lac en E.coli el cual codifica para el uso de lactosa como sustrato es regulado por un represor.
La atenuación (“attenuation”) regula la transcripción en bacterias al detectar razones de transcripción y traducción. Un ejemplo de esto es el operón para histidina

CONT. REGULACIÓN DE LA EXPRESIÓN GENÉTICA
Operón de Histidina:
• Cuando el suplido de histidina es adecuado, la traducción de la
proteína líder ocurre rápidamente. El pequeño pedazo de mRNA entre
la polimerasa de RNA y el primer ribosoma forma un “anillo atenuador”
que evita la transcripción de los subsecuentes genes en el operón.
Cuando el suplido de histidina es bajo, la traducción de la proteína líder
es baja. El pedazo grande de mRNA forma un anillo (loop)
“antiterminador” que evita la formación del anillo atenuador. Entonces
la transcripción de los genes del operón procede.
La traducción de las proteínas ribosomales codificadas por mRNA es
regulada por una proteína que tiene 2 funciones:
1) Incorporarse a un ribosoma
2) Inhibir la traducción
Cuando el suplido de proteínas ribosomales es correcto todas las
moléculas se incorporan a ribosomas. Cuando hay muchas proteínas
ribosomales libres, se activa la 2da función.

Attenuation (in genetics) is a proposed mechanism of
control in some bacterial operons which results in
premature termination of transcription and which is based
on the fact that, in bacteria, transcription and translation
proceed simultaneously. Attenuation involves a provisional
stop signal (attenuator), located in the DNA segment that
corresponds to the leader sequence of mRNA. During
attenuation, the ribosome becomes stalled (delayed) in the
attenuator region in the mRNA leader. Depending on the
metabolic conditions, the attenuator either stops
transcription at that point or allows read-through to the
structural gene part of the mRNA and synthesis of the
appropriate protein.

CAMBIOS EN LA INFORMACIÓN GENÉTICA DE LA CÉLULA
Genoma – es la suma total de de la información genética (DNA) de una
célula.
El genoma puede cambiar por: mutaciones o por intercambio genético.
En la mayoría de los genes procariotas están en su cromosoma. Algunos
están en moléculas circulares de DNA o plásmidos.
La mayoría de los procariotas tienen un cromosoma circular, pero algunos
tienen más de un cromosoma circular y algunos tienen cromosomas
lineales.
La mayoría de los plásmidos son circulares y muy pocos son lineales.
Los plásmidos codifican algunos aspectos no esenciales.
Plásmidos R codifican para la resistencia a antibióticos.
La mayoría de los eucariotas no poseen plásmidos y su genoma está
formado por pares de cromosomas.

MUTACIONES
Las mutaciones – son cambios en la secuencia de las bases de los
nucleótidos de un genoma.
Las mutaciones de punto – involucran un cambio en un par de nucleótidos
e incluyen:
Substituciones y 2 tipos de “frameshift mutations” , Inserciones y
Deleciones.
Las mutaciones se pueden categorizar por sus efectos:
a) Silent mutation
b) Missense mutation
c) Nonsense mutation
Agentes mutagénicos:
Agentes físicos o químicos que pueden aumentar la razón normal de
mutación.
Agentes físicos incluyen: radiación ionizante – rayos X y rayos gamma ;
radiación no-ionizante – rayos ultravioleta
Estos últimos pueden provocar la formación de dímeros de pirimidinas

MUTACIONES
Agentes mutagénicos químicos - incluyen a análogos de nucleótidos
Son químicos que se parecen estructuralmente a los nucleótidos y pueden
resultar en un pareo erróneo. Esto puede resultar en una inserción o
deleción y pueden terminar por causar un “frameshift mutation”.
Las células pueden reponer el DNA via “light repair” , “dark repair” ,
“base-excision repair” y “mismatch repair”. Si el daño es muy extenso se
recurre a una respuesta SOS.
Los investigadores han desarrollado métodos de distinguir células
mutantes de células wild-type. Estos métodos incluyen: positive
selection, negative (indirect) selection, Ames test. Este último se utiliza
para detectar agentes mutagénicos que puedan ser carcinógenos
potenciales.
