Aplicaciones industriales de las células enteras
Biotransformations using free enzymes as biocatalysts are now a fully accepted andvalidated synthetic methodology to prepare homochiral compounds used inpharmaceutical, food additives or agrochemical industries.
Contrarily to free enzyme Biocatalysis, the utility of whole cells as biocatalysts is wellknown although commercial applications have been largely restricted to special caseswhere the microorganism contains the enzyme required for a single step or where thecell is a natural producer of a chemical such as EtOH or a complex natural productsuch as tetracycline, astaxanthine etc.
This reduced utility has been associated to a lack of general and effective experimentaltools required to develop robust microorganism for industrial applications.
Nevertheless, the tremendous advances in the last years in metabolic engineering,genomics, metabolomics, proteomics, directed evolution etc. have opened the door tocreate tailored made microorganisms for industrial purposes.

• Due to the fact that HB27 strain grows in repetitive conditions and that its genome is sequenced, this strain was selected to get the enzymes.
6 genes were considered but only three showed good activity:
TTC0194: 2’-deoxypurine nucleoside synthesis.TTC1070: 2’-deoxypurine nucleoside synthesis. TTC1412: 2’-deoxypyrimidín nucleoside synthesis.
One pot nucleoside synthesis
The genes were cloned by standard methodology in an expression plasmid inducible by IPTG, and produced in the E. coli strain BL21(DE3).
Purification was carried out by heating at 80ºC, thus taking advance of the difference in optimal temperature of the host microorganism and the source microorganism.

Después se inserta la librería de genes mutados obtenidos en un microorganismo apropiado.Se expresan las enzimas mutadas y se prueban individualmente en la reacción de interés (enantioselectividad y conversión).

y se vuelve a repetir el proceso. Este proceso crea una presión evolutiva que lleva a la formación e identificación de la enzima de la enzima optima.

Empleando el método epPCR obtuvo 1.000 a 3.000 mutantes que utilizó en la hidrólisis del los esteres de p-nitrofenilo midiendo en el visible la aparición de p-nitro fenol empelando palcas de 96-pocillos lo que permite analizar entre 400 y 500 microorganismos día. Así obtuvo una lipasa mutada mejor que la cepa salvaje : e.e.81%, E=11.3.
Lipasa de P. aeruginosa en concreto

Empleando el método de barajeado a partir del menor gen obtuvo un gene mejorado con e.e.>95%, E > 51.

La tecnología microbiana presenta ventajas sobre los métodos no biológicos de recuperación de minerales, entre los que podemos
encontrar: 1)Requieren poca inversión de capital (las bacterias pueden ser aisladas a partir de aguas ácidas de minas).
Recuperación de minerales de minas pobres
2)Bajos costes de operación necesarios para las operaciones hidrometalúrgicas en comparación con los procesos convencionales. 3)Relativa ausencia de polución o contaminación ambiental durante el proceso. 4)El tratamiento del creciente cúmulo de minerales de baja ley en las minas que no pueden ser económicamente procesados por los métodos tradicionales.

The reduction is catalyzed by alcohol dehydrogenases (ADHs) (E.C. 1.1.1) which natural substratesare ethanol, lactate, glycerol etc.These enzymes require a coenzyme such as NADH or NADPH from which one hydride ion istransferred to C=O.There are four stereochemical patterns that enable the transfer of the hydride from the coenzymeto the substrate (Nakamura et al. 2003).E1 and E3 enzymes, the pro-(R)-hydride from NAD(P)H is transferred,E2 and E4 enzymes transfer the pro-(S)-hydride.
In E1 and in E2,hydride ion attacks the si- face but in E3 and E4 enzymes the attack is in re-face.

Enoate reductases (1.3.1.31) are NADH or NADPH dependent enzymes that reducesα,β-unsaturated ketones to saturated ketones. When an alkyl group is present in alpha position, the enzyme shows great stereoselectivity leading to the 2-R-stereomeras in the case of the reduction of carvones where (1R,4R)-dihydrocarvone or (1R,4S)- iso-dihydrocarvone are obtained from the 4R-carvone or 4S-carvone(Shimoda et al.1998).

Homochiral α-substituted ketones are versatile building blocks for the synthesisof pheromones (Mori and Ebata, 1986). Chemical methodologies carry to low e.e. values The reduction of α,β-disubstituted enones using whole cells such as baker’s yeast is a greenalternative because the reaction usually carries in aqueous medium with small amounts of hydrophilic solvents such as THF or 1,4-dioxane.
Kawai et al. (2001) described that the elongation of the α-substituent from methyl to ethylimproves the enantioselectivity. The substituent in the β-phenyl group is a critical factor. The reduction of enones having no-substituted or a para-substituent affords the (S)-α-substituted ketone with moderate stereoselectivity, whereas the presence of substituentin meta-position dramatically increases the enantioselectivity. Microbial Baeyer-Villiger oxidation is a valuable tool to obtain lactones from ketones in a stereoselective manThis biocatalysed reaction is a green alternative to the non stereoselective chemical process (Rudroff et al. 2007).
The process is catalyzed by a Baeyer-Villiger monoxygenase that is a flavin and NADH-dependent enzyme.Acinetobacter calcoaceticus cyclohexane monoxygenase (CMO) has been used as reference to optimizethe production of the enzyme by conventional Microbiological methodologies (Barclay et al. 2001) and to study the substrate specificity in the Baeyer-Villiger oxidation (Zambianchi et al. 2000) using racemic bicycle[3.2.0]hept-2-en-6-one as the substrate. Asymmetric hydroxylations of unfuntionalyzed hydrocarbons provide a powerful Method to obtain potentially useful, optically active oxyfunctionalized synthons of unactivated hydrocarbons.
